摘　　要：目的系统评价恩杂鲁胺用于前列腺癌安全性。方法通过检索并筛选网络在线数据PubMed、EMBASE、中国期刊全文数据库(CNKI)、万方数据库(从建库至2019年9月),收集其中关于恩杂鲁胺用于治疗前列腺癌的临床随机对照研究(RCT)。使用Revman5.3软件对纳入研究进行Meta分析。结果本次Meta分析共纳入4篇符合筛选标准的研究,共包含5455例患者资料。恩杂鲁胺组各级别疲劳、潮热、头痛、肌肉骨骼疼痛、高血压、虚弱乏力和心血管疾病的发生率高于安慰剂组,差异均有统计学意义(P<0.05)。恩杂鲁胺组患者各级别腹泻的发生率与安慰剂组比较,差异无统计学意义(P>0.05)。结论恩杂鲁胺治疗前列腺癌期间需要监护治疗疲劳、潮热、头痛、乏力虚弱、高血压、肌肉骨骼痛及心血管疾病等情况,腹泻的发生与恩杂鲁胺的使用无关。Objective To systematically evaluate the clinical safety of enzalutamide in the treatment of prostate cancer.Methods Weused online databases such as PubMed,EMBASE,CNKI and Wanfang Database to search randomized controlled trials(RCT)of enzalutamidein the treatment of prostate cancer published prior to September 2019.The Meta-analysis was conducted by using RevMan 5.3 software.Results Four independent studies with 5455 prostate cancer patients were included in the present study.The incidence of fatigue,hot flash,headache,musculoskeletal pain,hypertension,asthenia and adverse cardiovascular events of all gradesin the enzalutamide group was higher than those in the placebo group with statistic significance;The incidence of diarrhea of all grades in the enzalutamide group was higher than that in the placebo group but the difference was not statistically significant(P>0.05).Conclusion Prostate cancer patients treated with enzalutamide need to be monitored whether they may have fatigue,hot flashes,headache,asthenia,hypertension,musculoskeletal pain and cardiovascular disease.The incidence of diarrhea is not correlated with the drug use.
