摘  要:目的利用裸鼠动物模型,观察骨向分化过程中纤溶蛋白酶原激活剂抑制物-1(PAI-1)诱导人骨髓间充质干细胞(hBMSCs)骨向分化的能力。方法取拔牙时磨除的骨组织分离及培养hBMSCs,应用茜素红S染色检测向成骨细胞分化程度,并应用real-time PCR检测骨向分化的标志性基因(ALP、BSP、Col1、OCN、Run×2)的表达。然后,将hBMSCs移植到BALB/c裸鼠体内,采用组织学检查检测PAI-1在体内诱导hBMSCs形成骨组织能力。结果PAI-1不仅诱导hBMSCs向成骨细胞分化,还诱导ALP、BSP、Col1、OCN、Run×2基因的高表达。在体内PAI-1可诱导hBMSCs分化成骨组织,而且PAI-1诱导Col1和BSP在新生骨组织中的高表达。结论PAI-1可促进hBMSCs向成骨细胞分化及在体内诱导hBMSCs分化成骨组织。Objective To investigate the effect of plasminogen activator inhibitor-1(PAI-1)on inducing osteogenesis of human bone marrow stromal cells(hBMSCs).Methods Human BMSCs were harvested from human alveolar bone marrow and induced osteogenic differentiation with PAI-1.Alizarin red S(ARS)staining assay was used to assess osteogenic diffdrentiation of hBMSCs,and real-time PCR to evaluate the expression levels of osteogenic markers.H&E staining and immunohistochemistry were applied to detect PAI-1-induced new bone and the expression of Col1 and BSP.Results PAI-1 significantly promoted osteogenic differentiation of hBMSCs,showing high levels of mineralized nodule formation and expression of osteogenic markers.Furthermore,transplantation of hBMSCs with PAI-1 showed a great ability to induce bone formation.Conclusion PAI-1 promotes osteogenic differentiation of hBMSCs and has a strong effect on bone formation in hBMSCs.
