三种义齿稳固剂的pH值及其对变形链球菌生长的影响

摘　　要：目的体外研究3种商业化义齿稳固剂的pH值及其对变形链球菌生长的影响,探讨其应用于有天然牙存留患者的潜在致龋性。方法将适量稳固剂加入去离子水中,制成浓度分别为10.0%、5.0%、2.5%和1.25%(重量/体积比)(膏剂)或5.0%、2.5%、1.25%和0.625%(粉剂)的水溶液,分别在样品制备后0、1、2、3、6、12和24 h,使用数字化pH计测试各样品的pH值。在含有义齿稳固剂浸提液(实验组)或生理盐水溶液(对照组)的BHI液体培养基内接种厌氧培养变形链球菌,采用比浊法分别于12 h内每小时连续测定培养前后菌液的光密度(OD)值,以反映变形链球菌的生长情况。结果稳固剂的pH值在测试时间内(24 h)保持相对稳定,其范围为5.5~7.0。稳固剂浸提液处理组与对照组的变形链球菌生长率差异无统计学意义(P>0.05)。结论部分义齿稳固剂在特定浓度和时间点的pH值低于羟基磷灰石溶解的临界pH值,可能不适用于含天然牙的义齿佩戴者;所测义齿稳固剂在体外对变形链球菌生长无显著性影响。Objective To investigate the pH and effects of three commercial denture fixatives on Streptococcus mutans(S.mutans)growth.Methods Samples of each fixative were added to deionized water to produce solutions of 10.0%,5.0%,2.5%and 1.25%w/v or 5.0%,2.5%,1.25%and 0.625%w/v.The pH values were measured at 0,1,2,3,6,12 and 24h intervals using a digital pH meter.S.mutans UA159 was inoculated in the brain heart infusion medium with or without the fixative extracts(control).Nephelometer method was used to measure optical density(OD)at 600 nm of S.mutans culture fluid in the different incubation conditions every 1 h to 12 h using a spectrophotometer.Results The tested fixatives remained relatively stable pH value over 24 h,ranging from 5.5 to 7.0.There was no significant difference in S.mutans growth rates between the extract-treated and control cultures(P>0.05).Conclusion Certain fixatives produce a pH below the critical pH of hydroxyapatite and may not be suitable for patients with natural teeth.None of the tested fixatives significantly affects the growth of S.mutans.
