摘  要:目的分析单侧输尿管梗阻不同肾纤维化大鼠模型中肾组织肝细胞生长因子(HGF)、骨形态发生蛋白-7(MBP-7)、转化生长因子-β(TGF-β)、结缔组织生长因子(CTGF)的表达变化。方法选取温州医科大学实验动物中心提供的健康成年雄性清洁级大鼠64只,按照随机数字法进行分为假手术组、模型组、中药组、西药组,每组16只。给予4组大鼠不同的治疗方案并比较治疗后大鼠肾纤维化水平,对不同大鼠模型肾纤维化水平与肾组织HGF、MBP-7,TGF-β、CTGF的表达水平的相关性进行比较分析。结果4组大鼠第3、7、14天肾脏病理改变情况、HGF表达水平、TGF-β表达水平、BMP-7表达水平及CTGF表达水平差异有统计学意义(P<0.05)。肾纤维化程度与HGF、MBP-7及CTGF的表达水平呈正相关,与TGF-β的表达水平呈负相关。结论可通过HGF、MBP-7、TGF-β及CTGF的变化程度对肾纤维化程度进行判断。Objective To analyze the expression of hepatocyte growth factor(HGF),bone morphogenetic protein-7(MBP-7),transforming growth factor-β(TGF-β)and connective tissue growth factor(CTGF)in rat models with unilateral ureteral obstruction of different renal fibrosis.Methods A total of 64 healthy adult male rats were selected from the Experimental Animal Center of Wenzhou University and divided into the sham operation group,the model group,the Chinese traditional medicine group and the Western medicine group,according to random number method,with 16 rats in each group.Rats of 4 groups were given different treatment regimens.And levels of renal fibrosis after treatment were compared.The correlation between the level of renal fibrosis and the expression levels of HGF,MBP-7,TGF-βand CTGF in different rat models was analyzed.Results At the 3rd,7th and 14th day,pathological changes,HGF,TGF-βand CTGF expression among the 4 groups were statistically different(P<0.05).The degree of renal fibrosis was positively correlated with the expression levels of HGF,MBP-7 and CTGF,and negatively correlated with the expression level of TGF-β.Conclusion The degree of renal fibrosis can be judged by the changes of HGF,MBP-7,TGF-βand CTGF.
