摘  要:为了解广西PDCoV流行毒株在猪群中的感染和流行特点,用设计的25对特异性引物从腹泻样本中检测获得1株PDCoV全基因组序列。结果显示,CHN-GX01-2018株基因组全长25406bp,与美国毒株比较存在3处基因缺失,分别位于ORF1a/1b第400~403aa、第756~759aa位置及S基因存在1个氨基酸的缺失。全基因组与S基因遗传进化分析结果显示,CHN-GX01-2018与代表株CHN-AH-2004(安徽株)同源性为97.4%。然而,同源性分析显示中国PDCoV株也存在不同的进化分支,表明PDCoV在中国猪群中不断进化。Porcine deltacoronavirus(PDCoV)is a newly emerging enterotropic swine coronavirus that causes acute enteritis in nursing piglets.In order to understand the infection and epidemic characteristics of Guangxi PDCoV strains in pigs,25pairs of specific primers were designed to detect the full length of PDCoV genome from diarrhea samples.The results showed that the genome of CHN-GX01-2018strain is 25,406bp in length,and there are three gene deletions compared with the US strain:at 400-403aa,756-759aa in located at ORF1a/1b,and the Sgene has one amino acid deletion.Homology analysis showed that genetic analysis of whole genome and Sgene showed that CHN-GX01-2018had 97.4%similarity with CHN-AH-2004(Anhui strain).However,there are also different evolutionary branches of the Chinese PDCoV strain,indicating that PDCoV is constantly evolving in Chinese pig herds.This study plays a positive role in the epidemiological status of PDCoV in the herd and its genetic evolution,vaccine development,and disease prevention and control.
