摘 要:目的优化祁蛇肉酶解工艺,提高祁蛇多肽在水、酒等溶剂中的溶出率。方法通过多种酶解技术,将祁蛇中的大分子蛋白切割成多种小分子多肽,以祁蛇酶解后多肽峰面积为指标,对其酶解的工艺进行研究。结果祁蛇蛋白酶解优化的工艺为木瓜蛋白酶酶解加入量为3.5%,酶解时间为2.5 h;中性蛋白酶加入量3.5%,酶解时间为1.5 h;碱性蛋白酶加入量2.0%,酶解时间为1 h。结论运用该工艺能显著提高祁蛇多肽的溶出率,对进一步提高祁蛇的作用功效与利用率具有显著意义。Objective The dissolution rate of qi-snake polypeptides in water,liquor and other solvents was greatly increased.Methods Through a variety of enzymatic hydrolysis techniques,the large molecule of qi snake protein was cut into many small polypeptide molecules,The enzymatic hydrolysis of polypeptide was studied by using its peak area as an index.Results The best conditions for the protein hydrolysis of qi-snake were dosage of papain 3.5%,the enzymatic hydrolysis time was 2.5 hour;dosage of Neutral enzyme 3.5%,the enzymatic hydrolysis time was 1.5 hour;dosage of Alkaline enzyme 2.0%,the enzymatic hydrolysis time was 1 hour.Conclusion The polypeptide dissolution rate and the utilization rate of qi-snake can be improved by using this technology.
