摘　　要：目的检测T-BOX转录因子2(TBX2)在脑胶质瘤U251细胞中的表达情况,并探讨其对胶质瘤U251细胞生物行为学的影响。方法将体外培养的人脑胶质瘤细胞U251随机分成3组,转染含T-BOX转录因子2(TBX2)inhibitor慢病毒载体的TBX2 inhibitor组,转染携带无义序列的慢病毒载体的阴性对照组(NC组),及不转染的空白对照组(CK组),采用qRT-PCR法检测TBX2 mRNA相对表达量,采用CCK-8法检测细胞增殖情况,采用Transwell法检测细胞迁移与侵袭的情况。结果TBX2 mRNA在脑胶质瘤U251细胞中的表达水平显著高于正常细胞(P<0.05);TBX2 inhibitor组脑胶质瘤细胞TBX2相对表达量显著低于CK组、NC组(P<0.05);TBX2 inhibitor组脑胶质瘤细胞培养24 h、48 h、72 h的OD值均显著低于CK组、NC组(P<0.05);TBX2 inhibitor组脑胶质瘤细胞迁移数目显著低于CK组、NC组(P<0.05);TBX2 inhibitor组细胞侵袭数目显著低于CK组、NC组(P<0.05)。结论TBX2在脑胶质瘤细胞中表达上调,下调TBX2表达可抑制脑胶质瘤细胞的增殖、迁移与侵袭。Objective To detect the expression of TBX2 in gliomas cell U251 in vitro,and to explore its effects on the biological behavior of glioma cell U251.Methods The human glioma cell line U251 cultured in vitro was randomly divided into three groups,TBX2 inhibitor group[transfected with lentiviral vector containing T-BOX transcription factor 2(TBX2)inhibitor],negative control group(NC group,transfected with lentiviral vector carrying nonsense sequence),blank control group(CK group,non-transfected),the relative expression levels of TBX2 mRNA were detected by qRT-PCR,cell proliferation was detected by CCK-8 method,and the cell migration and invasion were detected by Transwell assay.Results The expression levels of TBX2 mRNA in glioma cells were significantly higher than those in normal cells(P<0.05),and the relative expression levels of TBX2 in TBX2 inhibitor group were significantly lower than those in CK group and NC group(P<0.05).The OD values at 24h,48h,and 72h after cell culture in TBX2 inhibitor group were significantly lower than those in CK group and NC group(P<0.05).Moreover the migration cell number in TBX2 inhibitor group was significantly lower than that in CK group and NC group(P<0.05),and invasion cell number in TBX2 inhibitor group was significantly lower than that in CK group and NC group(P<0.05).Conclusion TBX2 expression is up-regulated in gliomas cell,and down-regulation of TBX2 expression can inhibit proliferation,migration and invasion of glioma cells.
