摘 要:目的:探讨miR-21作用于人第10 号染色体缺失的磷酸酶及张力蛋白同源物(Phosphatase and tensin homology deleted on chromosome 10,PTEN)与结肠癌化疗疗效关系.方法:荧光定量PCR测定结肠癌组织中的miR-21、免疫组化检测PTEN的表达,化疗2周后进行疗效评估;培养结肠癌细胞株SW480组(对照组)、转染miR-21抑制剂抑制组以及化疗耐药组,qRT-PCR检测细胞miR-21的表达,Western blot 检测PTEN的表达,流式细胞术检测细胞凋亡.结果:miR-21在结肠癌组织中的表达明显高于癌旁正常组织(P<0.05),PTEN在结肠癌组织中表达减低(P<0.05),并且miR-21与PTEN的表达之间呈负相关(P<0.05);对患者进行化疗后疗效评估,发现化疗前组织 miR-21的表达在(CR+PR)组的表达量低于(PD+SD)组,而PTEN的表达在(CR+PR)组的表达量高于(PD+SD)组(P<0.05);三组细胞比较后发现各组细胞miR-21、PTEN的表达和细胞凋亡率都存在差异(P<0.001).结论:PTEN可作为miR-21调控的靶基因之一参与细胞的耐药过程,提示联合检测结肠癌中miR-21和PTEN可成为有效的辅助预测化疗疗效的敏感指标.Objective:To investigate the relationship between the effect of microRNA-21 on PTEN and the efficacy of chemotherapy in colon cancer.Methods:Real-time fluorescent quantitative PCR to detect the expression of miR-21,and immunohistochemistry to detect the expression of PTEN in these colon cancer tissues,then the curative effect was evaluated after two weeks of chemotherapy;we cultured three groups of cells:control group,transfection inhibition group,drug resistant cell group,then qRT-PCR was used to detect the expression of miR-21 in all groups of cells and western blot was used to detect the expression of PTEN,as well as flow cytometry to detect the apoptosis rate in each group.Results:The expression of miR-21 in stageⅢcolon cancer was significantly higher than adjacent normal tissues(P<0.05),the expression of PTEN in stageⅢcolon cancer was decreased(P<0.05),and the expression of miR-21 was negatively correlated with that of PTEN(P<0.05).The expression of miR-21 in(CR+PR)group was lower than that in(PD+SD)group,while the expression of PTEN in(CR+PR)group was higher than that in(PD+SD)group(P<0.05).After comparing the each group,we found that the expression of miR-21,PTEN and the apoptotic rate of each group were different(P<0.001).Conclusion:PTEN can be used as a target gene of miR-21 to participate in the process of cell resistance,suggesting that the combined detection of miR-21 and PTEN in colon cancer can be an effective and sensitive marker for adjuvant prediction of chemotherapy efficacy.
