摘 要:目的:通过对乙二醇灌胃的肾结石大鼠模型进行白藜芦醇干预,测定各组大鼠相关指标的相对浓度以及大鼠肾脏组织石蜡切片的病理变化及钙盐结晶的情况一起进行分析,研究其中可能存在的作用机制,希望能够对临床中肾结石的防治提供新的想法。方法:将购置来的40只康健SD雄性大鼠适应性喂养后,随机分为4组(A、B、C、D),每组10只,均正常饲料喂养,A组:饮用水,B组:1%乙二醇灌胃,C组:1%乙二醇+白藜芦醇(5mg/kg)灌胃,D组:1%乙二醇+白藜芦醇(10mg/kg)灌胃,于第五周处死大鼠,收集大鼠血液标本,检测大鼠血清肌酐、尿素氮;摘取大鼠肾脏检测肾组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量,并将肾组织行石蜡切片、HE染色,观察肾组织中晶体形成及肾小管损伤情况。结果:B组大鼠血清肌酐、尿素氮及肾组织中MDA酶的含量水平高于A组,而SOD酶活性低于A组(P<0.05);C、D二组被不同浓度的白藜芦醇干预后,测得二组大鼠血清肌酐、尿素氮及肾组织中MDA活性均低于B组,而SOD酶活性高于B组(P<0.05)。结论:白藜芦醇可能通过抗氧化应激作用对乙二醇所致大鼠肾结石的形成起到抑制作用,同时对大鼠的肾功能有一定保护作用。Objective:To explore the mechanism of resveratrol on glycol-induced kidney stones in rats,and to guide clinical diagnosis and treatment.Methods:Forty healthy SD male rats were randomly divided into 4 groups(A,B,C and D)after adaptive feeding.Group A:drinking water;group B:1%ethylene glycol;group C:1%ethylene glycol+resveratrol(5 mg/kg);group D:1%ethylene glycol+resveratrol(10 mg/kg);rats were sacrificed at the fifth week.Blood samples of rats were collected and serum creatinine and urea nitrogen were measured.The activity of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in the renal tissues of rats were detected,and the renal tissues were stained with paraffin sections and HE staining to observe the crystal formation and tubular damage in the renal tissues.Results:The content of MDA in serum creatinine,urea nitrogen and renal tissue of group B was higher than that of group A,while the activity of SOD was lower than that of group A(P<0.05).After the intervention with different concentrations of resveratrol in the C and D groups,the activity of MDA in serum creatinine,urea nitrogen and renal tissue of the two groups was lower than that of group B,while the activity of SOD was higher than that of group B(P<0.05).Conclusion:Resveratrol may inhibit the formation of kidney stones caused by ethylene glycol through anti-oxidative stress in rats,and has a protective effect on renal function in rats.
