摘  要:目的:探讨绿原酸对人骨肉瘤MG63细胞增殖、侵袭、迁移及凋亡的影响。方法:采用细胞计数试剂盒(CCK-8)检测细胞增殖,细胞侵袭实验(Transwell)及细胞划痕实验检测细胞侵袭性及迁移性,逆转录-聚合酶链反应(RT-PCR)检测细胞干性基因及凋亡相关基因,凋亡试剂盒检测细胞凋亡比例。结果:在100μg/mL绿原酸干预的实验组中MG63细胞增殖减慢,细胞干性基因较对照组表达降低,侵袭性及迁移性经绿原酸处理后也相应降低,抗凋亡基因Bcl-2表达下降,细胞凋亡比例增加。结论:绿原酸对MG63人骨肉瘤细胞的增殖、侵袭、迁移具有抑制作用并促进MG63细胞的凋亡,有望成为治疗骨肉瘤的有效药物。Objective:To investigate the effects of chlorogenic acid on the proliferation,invasion,migration and apoptosis of human osteosarcoma MG63 cells.Methods:Cell proliferation kit(CCK-8)was used to detect cell pro liferation.Cell invasion test(Transwell)and cell scratch test were used to detect invasiveness and migration of MG63 cells and reverse transcription-polymerase chain reaction(RT-PCR)was used to detect tumor stem cell genes and apoptosis-related genes.The apoptosis kit was used to detect the proportion of apoptosis.Results:In the experimental group(MG63 cells treated with 100μg/mL chlorogenic acid),the proliferation of MG63 cells was slowed down and the expression of cell stem genes was lower than that of the control group.The invasiveness and migration were also reduced after treated with chlorogenic acid.The expression of anti-apoptotic gene Bcl-2 gene decreased while the proportion of apoptosis increased in the experimental group.Conclusion:Chlorogenic ac id can inhibit the proliferation,invasion and migration of MG63 human osteosarcoma cells and promote the apopto sis of MG63 cells.It is expected to be an effective drug for treating osteosarcoma.
