摘  要:目的探讨卵巢良、恶性肿瘤组织中人端粒酶逆转录酶(hTERT)启动子DNA甲基化的表达差异。方法收集15例卵巢恶性肿瘤患者术中新鲜肿瘤组织标本,设为实验组;另取15例卵巢良性肿瘤患者术中新鲜肿瘤组织标本,设为对照组;培养Hela细胞3组,作为实验阳性结果对照组。所有实验组织及细胞标本均提取DNA,进行甲基化特异性聚合酶链式反应,以Hela细胞甲基化结果作为阳性对照组,比较3组hTERT启动子DNA甲基化阳性率。结果实验组甲基化阳性率为73.3%(11/15),对照组甲基化阳性率为26.7%(4/15),阳性对照组甲基化阳性率为100.0%(3/3)。实验组甲基化阳性率明显高于对照组,差异有统计学意义(P=0.027)。结论与卵巢良性肿瘤组织比较,卵巢恶性肿瘤组织hTERT启动子DNA甲基化率明显增高。Objective To investigate the difference of methylation status of human telomere enzyme reverse transcriptase(hTERT)gene promoter DNA between the benign and malignant ovarian tumor.Methods 15 patients with malignant ovarian tumors(experiment group)and 15 patients with benign ovarian neoplastic cysts(control group)confirmed by pathological examination were recruited from Hangzhou Women's Hospital.The methylation of hTERT promoter DNA was detected by methylation-specific PCR(MSP),taking cultured Hela cell as positive reference.The DNA methylation rate of hTERT promoter in two groups were compared.Results MSP analysis showed that the positive methylation rate in experiment group was 73.3%(11/15),which was significantly higher than that in control group(26.7%,4/15,P=0.027),the methylation rate in Hela cells was 100.0%.Conclusion Compared with benign ovarian tumors,ovarian malignant tumors have a higher methylation rate of hTERT promoter DNA.
