雷替曲塞对人结肠癌细胞SW480增殖、凋亡的影响及其机制

摘　　要：目的观察雷替曲塞对人结肠癌细胞(SW480)细胞增殖及细胞凋亡的影响,并探讨其机制。方法将SW480随机分为A组、B组、C组与对照组,A组加入雷替曲塞0.1μg/mL,B组加入雷替曲塞0.5μg/mL,C组加入雷替曲塞2.5μg/mL,对照组加入等量培养液。培养24、48、72 h后采用CCK-8试剂盒检测各组SW480增殖活性;培养10 d后,计算各组SW480克隆形成率;培养72 h后,采用实时荧光定量PCR检测Bax、Bcl-2及Caspase-3 mRNA,采用Western Blotting法检测Bax、Bcl-2及Caspase-3蛋白。结果随着雷替曲塞浓度的增加以及干预时间的延长,SW480的增殖速度减慢(P均<0.05),且在浓度达到一定程度后,该现象不再明显;孵育10 d后,各组SW480克隆形成率组间两两比较,P均<0.05;随着雷替曲塞浓度的增加,SW480 Bax及Caspase-3蛋白及mRNA表达升高,Bcl-2蛋白及RNA表达降低(P均<0.05)。结论雷替曲塞可通过调节SW480 Bax、Caspase-3和Bcl-2蛋白及其RNA的表达,抑制细胞增殖,诱导结肠癌细胞凋亡。Objective To investigate the effects of raltitrexed on the cell proliferation and apoptosis of human colon cancer cells(SW480)and to in vestigate its mechanism.Methods SW480 cells were randomly divided into the group A,group B,group C and the control group.Cells in the group A were added with 0.1μg/mL raltitrexed,group B with 0.5μg/mL raltitrexed,group C with 2.5μg/mL raltitrexed,and the control group with an equal amount of culture medium.After 24,48,and 72 hours of culture,the proliferation activity of SW480 cells in each group was detected by using CCK-8 kit;after 10 days of culture,the colony formation rate of SW480 cells in each group was calculated;after 72 h of culture,Bcl-2 assaciated X protein(Bax),B-cell lymphoma-2(Bcl-2)and Caspase-3 mRNA were detected by real-time fluorescence quantitative PCR(qRT-PCR);Bax,Bcl-2 and Caspase-3 proteins were detected by Western Blotting.Results Over the time of treatment and with the increased concentrations of raltitrexed,the proliferation rate of SW480 cells slowed down(all P<0.05),and after the concentration reached a certain level,the phenomenon was no longer obvious;after 10 days of incubation,significant difference was found in the the formation rate of SW480 clones between groups(all P<0.05);as the concentration of raltitrexed increased,the expression of SW480 Bax and Caspase-3 protein and RNA increased,and the expression of Bcl-2 protein and RNA decreased(all P<0.05).Conclusion Raltitrexed can inhibit the cell proliferation and induce apoptosis of colon cancer cells by regulating the expression of SW480 Bax,Caspase-3 and Bcl-2 proteins and their mRNAs.
