摘 要:为了比较HPD826氢键型吸附树脂比通用型D101树脂对极性较强的原花青素有较好的纯化作用,且采用树脂法制备的提取物是否具有确切的内皮依赖性舒张动脉血管的活性,本研究通过静态试验比较了2种大孔吸附树脂对山楂果中原花青素的吸附-解吸性能,HPD826型吸附量、解析率及吸附速率常数均优于D101型。用HPD826树脂通过动态试验优化了吸附分离程序,上样浓度为25 mg/mL,流速为1.5 BV/h,解吸程序为:水洗1.5BV;45%(v/v)乙醇解吸1.5 BV;90%乙醇活化洗脱1 BV;最后水洗2 BV,洗脱流速为1.5 BV/h。用这种程序,45%乙醇解吸部分的原花青素得率为81.48%,经HPLC测定,制备的提取物中以表儿茶素和原花青素B2为主,10种原花青素成分总含量达干重的42%,其对内皮完整的血管具有舒张作用,浓度在1.4~140 mg/L范围内。The research aims to verify that HPD826 hydrogen-bonded adsorption resin has a better purification effect on the proanthocyanidins with strong polarity than general-purpose D101 resin,and make sure whether the extract prepared by the resin method has exact endothelial-dependent relaxation activity of arterial vessels.The adsorptiondesorption properties of procyanidin from hawthorn by two macroporous adsorption resins were compared by static experiments.The adsorption,resolution and adsorption rate constants of HPD826 were better than those of D101.The adsorption separation procedure was optimized by dynamic test with HPD826 resin.The loading concentration was 25 mg/mL,and the flow rate was 1.5 BV/h.The desorption procedure was 1.5 BV for washing,45%(v/v)ethanol desorption for 1.5 BV,90% ethanol activation eluting 1 BV,and finally washing 2 BV with an elution flow rate of 1.5 BV/h.Using this procedure,the yield of procyanidin was 81.48%in the 45% ethanol desorption fraction.The extracts prepared by HPLC were mainly epicatechin and procyanidin B2,and the total content of 10 procyanidin was 42%of the dry weight.It has a concentration-dependent relaxation effect on the rat isolated thoracic aorta ring precontracted by PE in the range of 1.4-140 mg/L.
