摘 要:帕金森病(PD)是一种常见的神经系统退行性疾病且不能治愈,其最主要的病理改变为中脑黑质多巴胺能神经元的变性死亡及路易小体的形成,进而引起纹状体内多巴胺显著减少。但导致其发病的具体分子机制尚未明确。Abelson酪氨酸激酶(c-Abl)是一种非受体酪氨酸激酶,其参与信号转导、DNA损伤修复、细胞凋亡周期调控、基因转录调控等重要的细胞活动进程。PD患者死后大脑中的c-Abl水平和活性升高,且c-Abl抑制剂对PD存在保护作用。因此,对c-Abl与PD关系的进一步研究有助于深入了解PD的发生和发展,为PD的临床防治提供新方向。Parkinson′s disease(PD)is a common neurodegenerative disease and cannot be cured.The most important pathological change is the degeneration of dopaminergic neurons in the midbrain and the formation of Lewy bodies,causing a significant reduction in striatal dopamine content.However,the specific molecular mechanism leading to its pathogenesis has not yet been clarified.Abelson tyrosine kinase(c-Abl)is a non-receptor tyrosine kinase involved in important cellular processes including signal transduction,DNA damage repair,cell cycle regulation,gene transcription regulation,etc.The level and activity of c-Abl in the brain of PD patients increases after death,and c-Abl inhibitor has protective effect on PD.Therefore,the relationship between c-Abl and PD should be further studied in the future to help to understand the occurrence and development of PD,thus providing a new direction for the clinical prevention and treatment.
