金刚藤对人肝癌SMMC-7721细胞增殖、凋亡及细胞周期的影响

摘　　要：目的观察金刚藤对人肝癌SMMC-7721细胞增殖、凋亡及细胞周期的影响。方法对SD大鼠分别以0.5、1、2 g/mL的金刚藤水煎液灌胃,获得含药血清。将SMMC-7721细胞分为Control组、JGT-L组、JGT-M组、JGT-H组,分别给予生理盐水和0.5、1、2 g/mL的含药血清4 mL。继续培养12、24、48 h后采用CCK-8法检测细胞增殖能力。培养24 h后采用流式细胞术检测细胞凋亡情况及细胞周期分布。结果各金刚藤药物组相同作用时间下细胞增殖能力均低于Control组(P均<0.05);JGT-L组、JGT-M组、JGT-H组细胞增殖能力逐渐减弱,组间两两相比,P均<0.05。各金刚藤药物组细胞凋亡率均高于Control组(P均<0.05);JGT-L组、JGT-M组、JGT-H组细胞凋亡率逐渐升高,两两相比,P均<0.05。JGT-M组、JGT-H组G 0/G 1期细胞比例高于Control组(P均<0.05);JGT-M组、JGT-H组S期细胞比例低于JGT-L组,JGT-H组S期细胞比例低于Control组(P均<0.05);JGT-L组、JGT-M组G 2/M期细胞比例低于Control组(P均<0.05)。结论金刚藤可抑制SMMC-7721细胞增殖、诱导细胞凋亡,并将细胞周期阻滞于G 0/G 1期。Objective To observe the effects of Smilax china L.on the proliferation,apoptosis and cell cycle of human liver cancer SMMC-7721 cells.Methods SD rats were administrated 0.5,1,and 2 g/mL Smilax china L.decoction,respectively,to obtain drug-containing serum.SMMC-7721 cells were divided into the Control group,JGT-L group,JGT-M group,and JGT-H group,and 4 mL of normal saline,0.5,1,and 2 g/mL drug-containing serum were administered.After 12,24,and 48 hours of culture,CCK-8 was used to detect the cell proliferation.After 24 hours of culture,apoptosis and cell cycle distribution were detected by flow cytometry.Results The cell proliferation ability of each Smilax china L.drug group was lower than that of the Control group under the same action time(all P<0.05);the cell proliferation abilities of the JGT-L group,JGT-M group,and JGT-H group decraesed gradually,with statistically significant difference(all P<0.05).The apoptotic rate of each Smilax china L.drug group was higher than that of the Control group(all P<0.05);the apoptotic rates of the JGT-L group,JGT-M group,and JGT-H group gradually increased(all P<0.05).The proportion of cells in the G 0/G 1 phase of the JGT-M group and the JGT-H group was higher than that in the Control group(both P<0.05);the proportion of S-phase cells in the JGT-M and JGT-H groups was lower than that in the JGT-L group and the JGT-H group.The proportion of cells in the S phase was lower than that in the Control group(P<0.05);the proportions of cells in the G 2/M phase of the JGT-L group and JGT-M group were lower than that in the Control group(all P<0.05).Conclusion Smilax china L.can inhibit the proliferation and induce apoptosis of MMC-7721 cells,and block the cell cycle in G 0/G 1 phase.
