摘  要:目的比较颅骨源性间充质干细胞(cMSCs)移植与骨髓间充质干细胞(bMSCs)移植对缺血性脑卒中大鼠模型的治疗作用。方法从雄性SD大鼠的股骨和胫骨中分离出bMSCs,从颅骨中分离cMSCs,采用qRT-PCR法检测细胞中神经嵴相关基因(Snail、Slug)和神经营养因子(Bdnf、Gdnf、Ngf)的mRNA。将SD雄性大鼠建立大脑中动脉闭塞(MCAO)模型,另选20只为假手术组,将模型建立成功的大鼠随机分为模型组、bMSCs组和cMSCs组,各20只。模型组单纯给予PBS,bMSCs组和cMSCs组大鼠分别在MCAO后24 h经尾静脉注射bMSCs、cMSCs(1.0×10^6/300μL PBS)。分别于建模后2、7、14、21 d,采用平衡木实验和Garcia评分评价各组大鼠的运动功能;建模21 d后处死大鼠并取出大脑、制片,HE染色观察大鼠海马组织病理变化,采用TTC染色观察并计算大鼠海马组织局灶性梗死(CI)面积,采用TUNEL染色观察大鼠海马神经元凋亡情况并计算凋亡率。结果cMSCs中Snail、Slug、Bdnf、Ngf mRNA相对表达量均高于bMSCs(P均<0.05)。模型组、bMSCs组、cMSCs组在各时点平衡木实验得分高于假手术组,Garcia评分低于假手术组(P均<0.05)。bMSCs组、cMSCs组建模后7、14、21 d的平衡木实验得分低于模型组,Garcia评分高于模型组(P均<0.05);cMSCs组建模后7、14、21 d平衡木实验得分低于bMSCs组,Garcia评分高于bMSCs组(P均<0.05)。建模21 d后,与bMSCs组相比,cMSCs组的损伤相对减轻,坏死区域缩小,细胞结构不完整,核固缩程度下降,间质水肿减轻。bMSCs组、cMSCs组CI面积小于模型组,cMSCs组CI面积小于bMSCs组(P均<0.05)。模型组、bMSCs组、cMSCs组海马组织中神经元凋亡率高于假手术组,bMSCs组、cMSCs组神经元凋亡率低于模型组,cMSCs组神经元凋亡率低于bMSCs组(P均<0.05)。结论cMSCs来源于颅神经嵴细胞,具有显著的神经源性活性。cMSCs移植较bMSCs移植更有利于缺血性脑卒中大鼠运动功能恢复,可减轻海马组织损伤,减少海�Objective To compare the therapeutic effect of transplantation of cranial mesenchymal stem cells(cMSCs)with that of bone marrow stem cells(bMSCs)on ischemic stroke rats.Methods The bMSCs were isolated from the femur and tibia of male SD rats and cMSCs were isolated from the skull.Real-time PCR was used to detect the mRNA expression of neural crest-related genes(Snail,Slug)and neurotrophic factors(Bdnf,Gdnf,Ngf).The middle cerebral artery occlusion(MCAO)models were established in SD male rats.Another 20 rats were selected as the sham operation group.The MCAO model rats were randomly divided into the model group,BMSCs group and CMSCs group,with 20 rats in each.The rats in the BMSCs group and CMSCs group were injected with bMSCs and cMSCs(1.0×10^6/300μL PBS)via caudal vein at 24 h after MCAO.On the 2nd,7th,14th and 21st days after modeling,balance beam test and Garcia score were used to evaluate the motor function of rats in each group.After 21 days of modeling,rats were killed and their brains were taken out.HE staining was used to observe the pathological changes of rat hippocampal tissues.Triphenyltetrazolium chloride(TTC)staining was utilized for the calculation of cerebral infarction(CI)area,and and TUNEL staining for hippocampal neuron apoptosis as well as the apoptosis rate.Results The expression levels of Snail,Slug,Bdnf,and Ngf mRNA in cMSCs were significantly higher than those in bMSCs(all P<0.05).The experimental scores of balance beam in the model group,BMSCs group and CMSCs group were higher than those in the sham operation group,and Garcia score was lower than that in the sham operation group(all P<0.05).Compared with the model group,the experimental scores of balance beam in the bMSCs group and cMSCs group decreased significantly on the 7th,14th and 21st days after modeling,while the Garcia score increased significantly(all P<0.05),and the improvement degree of the cMSCs group at each time point was significantly higher than that of bMSCs group(all P<0.05).At 21st day after modeling,compared with th
