摘 要:目的:研究原位反应法制备磁性脂质体的最优工艺。方法:采用原位反应法制备磁性脂质体,以磷脂含量为评价指标,采取单因素试验和Box-Behnken响应面设计,优化制备工艺。结果:磁性脂质体制备的最优条件为磷脂0.75 g,无水乙醇用量70 mL,老化时间50 min,此时磁性脂质体中磷脂含量为146.79μg/mg。结论:本研究结合单因素试验和Box-Behnken响应面设计优化了原位反应法制备磁性脂质体的工艺条件,所得的工艺重现性好,稳定性好。Objective: To optimize the preparation of magnetic liposomes by in situ reaction method. Methods: The magnetic liposomes were prepared by in situ reaction method. The preparation process was optimized by combining single factor experiments and Box-Behnken response surface design. Results: The optimal conditions of preparing magnetic liposomes were that lecithin was 0.75 g, ethanol was 70 mL and aging time was 50 min. Under such conditions, the lipid content was 146.79 μg/mg. Conclusion: The preparation of magnetic liposomes by in situ reaction method was optimized in this research. And the optimized technology is stable and reliable.
