天智颗粒对慢性脑缺血大鼠脑组织Drebrin表达的影响

摘　　要：目的观察天智颗粒对慢性脑缺血大鼠Drebrin表达的影响。方法采用永久性双侧颈总动脉结扎制备慢性脑缺血模型,随机分为正常对照组、假手术组、模型组和天智颗粒治疗组,治疗组为造模成功后第2天开始进行天智颗粒以每天5 g/kg剂量灌胃,4周后通过水迷宫实验评价大鼠学习记忆能力,采用免疫组化法观察各组脑组织的Drebrin的表达。结果模型组大鼠的记忆力显著下降(P<0.05),治疗组较模型组明显好转(P<0.05)。模型组与假手术组相比Drebrin阳性细胞数明显减少(P<0.05),治疗组Drebrin阳性细胞数与假手术组相比明显减少(P<0.05),与模型组相比增多(P<0.05)。结论天智颗粒可能通过上调Drebrin的表达,调节树突棘可塑性,进而改善慢性脑缺血所致的脑功能障碍。Objective To study the effects of tianzhi granules on the expression of Drebrin by the chronic hypoperfusion model. Methods The model of chronic ischemic was the ermanent occlusion of the bilateral common carotid arteries(2VO) model.The rats were divided randomly into four groups:the normal control group,the sham-operated group,the model group and the treatment group.At the second day after the operation,the rats of the treatment group were given tianzhi granules(every day 5 g/kg) by intragastric administration.After 4 weeks,Morris water maze would be used to detect the function of rats’ learning and memory and observe the expression of Drebrin in rats brain by immunohistochemistry. Results Compared with the sham-operated group,the escape latency of the model group was prolongation obviously(P<0.05),and the escape latency of the treatment group was shorten compared with the model group(P <0.05). Compared with the sham-operated group,the numbers of Drebrin-positived cells in the model group was obviously decreased(P <0.05),the numbers of Drebrin-positived cells in the treatment group was decreased obviously compared to the sham-operated group(P<0.05),though it was increased compared to the model group(P <0.05). Conclusion Tianzhi granules may regulate dendritic spine plasticity by up regulating Drebrin expression,and then improve brain dysfunction caused by chronic cerebral ischemia.
