摘 要:目的:利用Ca-antimiR138进行钛种植体表面修饰以提升其表面骨诱导能力。方法:纯钛片抛光后在氢氟酸中进行阳极氧化(20 V,30 min)制备纳米管表面,将Ca-antimiR138复合物吸附于纳米管表面,将间充质干细胞(MSCs)培养在种植体表面并诱导成骨分化,7 d后采用BCIP/NBT试剂盒进行碱性磷酸酶(ALP)染色。结果:Ca-antimiR138复合物可高效吸附在纳米管表面,呈现均匀分布;采用Ca-antimiR138修饰后表面诱导ALP表达提升至10倍以上。结论:应用Ca-antimiR138复合物修饰种植体表面可显著提升其成骨诱导活性。Objective:To immobilize the Ca-antimiR138 complex onto titanium implant surface for the enhancement of osteogenesis(MSCs)of mesenchymal stem cells.Methods:Pure titanium sample surface was polished and anodized in hydrofluoric acid under 20 V for 30 min to formulate the nanotubes(NTs)structure.The Ca-antimiR138 complex was adsorbed and MSCs were seeded onto the NTs surface for the osteogenic differentiation of MSCs.After 7 days induction,the alkaline phosphatase(ALP)activity was determined by BCIP/NBT staining.Results:The Ca-antimiR138 complex was efficiently adsorbed onto NTs surface and showed relative homogeneous distribution.After the antimiR138 was loaded,the ALP activity of MSCs was increased by more than 10 times.Conclusion:The Ca-antimiR138 modified titanium implant surface can improve the osteoinductive ability of titanium implant.
