摘  要:目的研究端粒保护蛋白-1(POT1)和端粒结合因子-1(TRF1)在侵袭性垂体瘤与非侵袭性垂体瘤的表达水平及其临床意义。方法收集42例诊断明确、经手术治疗的垂体瘤患者,其中侵袭性垂体瘤12例、非侵袭性垂体瘤30例。分别应用实时荧光定量PCR及Western Blot法,检测手术切除的垂体瘤组织中POT1、TRF1基因和蛋白的表达水平,并探讨其与垂体瘤侵袭性的关系。结果侵袭性垂体瘤组的POT1、TRF1 mRNA表达水平显著高于非侵袭性垂体瘤组,而侵袭性垂体瘤组的POT1及TRF1蛋白表达水平同样显著高于非侵袭性垂体瘤组,差异均有统计学意义(均P<0.01)。巨腺瘤的POT1和TRF1 mRNA表达量显著高于微腺瘤,差异有统计学意义(P=0.0075,P=0.005)。结论POT1和TRF1在侵袭性垂体瘤及巨腺瘤中的表达水平显著增高。TRF1和POT1可能与侵袭性垂体瘤的发生、发展有密切关系。Objective To detect the expression of telomere-protected protein-1(POT1)and telomere repeat binding factor-1(TRF1)in invasive pituitary adenomas and non-invasive pituitary adenomas,and to explore its clinical significance.Methods A total of 42 well-diagnosed pituitary tumor specimens were collected,including 12 invasive pituitary adenomas and 30 non-invasive pituitary adenomas.Reverse Transcription-Polymerase Chain Reaction(RT-PCR)and Western blot were performed to assay the expression of telomere-protected protein-1(POT1)and telomere repeat binding factor-1(TRF1)in pituitary adenomas tissues.And the relationship between the expression level and the invasiveness of pituitary tumors was explored.Results The expression of POT1 and TRF1 in invasive pituitary adenomas were higher than those in non-invasive pituitary adenomas and the difference was significant(P<0.05,n=42).The expression levels of POT1 and TRF1 proteins in the invasive pituitary tumor group were also significantly higher than those in the non-invasive pituitary tumor group(P<0.01).The expression of POT1 and TRF1 mRNA in giant adenoma was significantly higher than that in micro adenoma,and the difference was statistically significant(P=0.0075,P=0.005).Conclusion The expression of POT1 and TRF1 in invasive pituitary adenomas tissues is significantly higher than that in non-invasive pituitary adenomas,suggesting that TRF1 and POT1 may be closely related to the occurrence and development of invasive pituitary adenomas.
