摘 要:目的研究制何首乌提取物对创伤后应激障碍(PTSD)大鼠神经保护作用及其机制。方法60只雄性SD大鼠随机分为5组,每组各12只。对照组大鼠常规饲养;PTSD模型组大鼠以幽闭电击法建立PTSD模型;低、中、高剂量制何首乌组大鼠造模后分别予以0.5、1.5、3 g/kg制何首乌提取物腹腔注射。比较各组大鼠的Morris水迷宫(MWM)评分、神经损伤严重缺损评分(NSS)、旷场试验(FT)、高架十字试验(ECT)结果、海马区细胞形态学、海马区神经元细胞凋亡灰度值、核因子Kappa B/抑制蛋白(NF-κB/IκB)通路p65、pIκBα/IκBα表达水平。并在给药大鼠中挑选改善效果最好的剂量组,给予NF-κB激活剂CHPG,观察其对海马区神经元凋亡的影响。结果高、中、低剂量组大鼠逃避潜伏期、NSS评分低于PTSD模型组,穿越平台位置次数、在原平台所在象限的时间占比、中央进入次数、水平活动度、开臂停留时间、开臂进入次数高于PTSD模型组。并且3组中剂量越高组大鼠以上指标改善越明显(均P<0.05)。PTSD模型组大鼠海马细胞排列疏松、胞浆染色深浅不一、细胞数目减少、体积缩小,存在固缩、变性神经元。低、中、高剂量组较模型组依次改善;高、中、低剂量组大鼠海马区细胞凋亡灰度值低于PTSD模型组;并且3组中剂量越高组大鼠的海马区细胞凋亡灰度值越低(均P<0.05)。高、中、低剂量组p65、pIκBα/IκBα低于PTSD模型组,且3组中剂量越高组的p65、pIκBα/IκBα表达降低幅度越大(均P<0.05)。应用CHPG大鼠海马区细胞凋亡灰度值显著高于未应用CHPG大鼠(均P<0.05)。结论制何首乌提取物可改善PTSD大鼠行为学和海马区损害,并能呈剂量依赖性靶向NF-κB/IκB通路调控海马区神经元凋亡。Objective To study the neuroprotective effect of Polygonum multiflorum extract on post-traumatic stress disorder(PTSD)rats and its mechanism.Methods A total of 60 male SD rats were randomly divided into 5 groups,12 cases in each group.Control group rats were routinely reared.PTSD model group rats established PTSD model by claustrophobic shock method.After low,medium and high-dose rats of Polygonum multiflorum were modeled,0.5,1.5,and 3 g/kg of Polygonum multiflorum extract was injected intraperitoneally.The Morris water maze(MWM)score,neurological injury severe defect score(NSS),open field test(FT),elevated cross test(ECT)results,hippocampal cell morphology,hippocampal neuron cell apoptosis gray value,nuclear factor Kappa B/Inhibitory protein(NF-κB/IκB)pathway p65,pIκBα/IκBαexpression levels in each group were compared.And the dose group with the best improvement effect was selected in the administered rats,and the NF-κB activator CHPG was given.The effect on neuron apoptosis in hippocampus was observed.Results The escape latency and NSS score of rats in the high,medium and low dose groups were lower than those in the PTSD model group,the number of times of crossing platform position,the proportion of time in the quadrant where the original platform was,the number of central entry,horizontal activity,the times of arm opening stay and the times of arm opening entry were higher than those of PTSD model group.And the higher the dose in the three groups,the more significant improvement in the above indicators of the rats(all P<0.05).In the PTSD model group,the hippocampal cells were loosely arranged,the cytoplasm stained differently,the number of cells was reduced,and the volume was reduced,and there were shrinkage and degeneration neurons.The low,medium,and high dose groups improved sequentially in comparison with the model group.The gray value of apoptosis in the hippocampus of rats in the high,middle and low dose groups was lower than that in the PTSD model group.And the higher the dose in the three groups,
