摘　　要：目的以胃蛋白酶原料为研究对象,通过光密度定量蛋白酶谱法分析胃蛋白酶的功效。方法以Native-PAGE负染电泳以及光密度数据分析技术为依托,进行方法学验证,利用该方法对胃蛋白酶进行分析研究,对产品质量及功效直观评价。结果建立了可以直观表征及定量比较酶功效的电泳技术,通过光密度数据提取对4种胃蛋白酶原料功效进行比较,与传统效价测定方法比较,结果更加精确,能够直观反映酶制剂的比酶活信息。结论该法测定结果反映了实际活性成分,能够更加直接反映胃蛋白酶的功效及质量,具有较好的应用前景,该方法亦可应用在其他酶制剂的分析研究中。Objective To study the efficacy of pepsin in pharmaceutical raw materials by optical density densitometry protease spectrometry with raw materials as the research object.Methods The method based on Native-PAGE negative dye electrophoresis and optical density data analysis was validated.The pepsin material were analyzed and evaluated,and the product quality and efficacy were evaluated visually.Results The electrophoresis technique was established to directly characterize and quantify enzyme activity.By comparing the efficacy of protease components in four kinds of pepsin raw materials by optical density data extraction,the result was more accurate and can directly reflect the specific enzyme activity information of the enzyme preparation.Conclusion The results of this method reflected the actual active ingredients in the sample,which can directly reflect the efficacy and quality of pepsin in the sample.The method had good application prospect and can be applied in the analysis of other enzyme preparations.
