摘 要:为研究复方黄芪降糖颗粒(FFHQ)对高血糖模型动物的降血糖作用及其机制,通过尾静脉注射四氧嘧啶(ALX)致糖尿病小鼠模型和体外HepG2细胞胰岛素抵抗试验,考察空腹血糖(GLU)和糖耐量(GT)等指标,对FFHQ降血糖功能性进行评价,同时测定血清胰岛素、肝糖原和肌糖原水平及HepG2细胞胰岛素抵抗情况,对其作用机制进行初步探讨。结果表明,FFHQ经给药30 d后,高、中和低剂量组均能显著降低糖尿病小鼠的GLU、提升GT(P<0.05或0.01),具有较好的降血糖作用。高剂量的FFHQ对正常小鼠的血糖水平无显著影响。通过对FFHQ作用机制的研究发现,该复方具有促进小鼠胰岛素分泌、增加肝糖原和肌糖原含量、改善胰岛素抵抗的作用。In order to study the hypoglycemic effect and mechanism of compound astragalus hypoglycemic granules on hyperglycemic model animals,diabetic mice model induced by tail vein injection of alloxan(ALX)and insulin resistance experiment of HepG2 cells in vitro were used to investigate the indexes of fasting blood glucose(GLU)and glucose tolerance(GT),so as to evaluate the hypoglycemic function of compound astragalus hypoglycemic granules(FFHQ)and determine serum insulin,liver glycogen and muscle glycogen levels and insulin resistance of HepG2 cells.The mechanism of action was discussed.The results showed that the high,middle and low dose groups of FFHQ could significantly reduce the GLU and increase the GT of diabetic mice(P<0.05 or 0.01),and had a better hypoglycemic effect.High dose FFHQ had no significant effect on the blood glucose level of normal mice.Through the study on the mechanism of FFHQ,it was found that the compound could promote the secretion of insulin,increase the content of glycogen in liver and muscle,and improve insulin resistance.
