摘 要:目的:探讨STAT3蛋白的表达与甲状腺癌发病的关系。方法:应用计算机检索截至2018年9月10日PubMed、Embase、CNKI和万方数据库中关于STAT3蛋白水平与甲状腺癌关系的病例对照研究。按照纳入和排除标准选择文献,提取相关的数据,以NOS量表评价其方法学质量,采用Stata 12.0软件对纳入文献进行统计学分析,异质性检验采用I2检验。结果:共纳入文献9篇,总样本量为711例。与对照组比较,甲状腺癌患者样本中STAT3蛋白水平明显增加,差异有统计学意义(RR=3.366,95%CI:2.657~4.264,P<0.05)。对甲状腺癌组内的样本分组进行分析,在有淋巴结转移的甲状腺癌组织中STAT3的表达比无淋巴结转移的甲状腺癌组织高(RR=1.313,95%CI:1.149~1.500,P<0.05);在TNF分期为Ⅲ~Ⅳ期的甲状腺癌组织表达高于Ⅰ~Ⅱ期甲状腺癌组织(RR=0.701,95%CI:0.558~0.882,P<0.05)。结论:STAT3蛋白在甲状腺癌组织中高表达,在甲状腺癌的发生发展中有重要的作用,有望成为甲状腺癌的一个新的治疗靶点。Objective:To explore the relationship between the expression of STAT3 protein and the pathogenesis of thyroid cancer.Methods:PubMed,Embase,CNKI and Wanfang database were searched from the establishment of the database to September 10,2018.A case-control study to retrieve the relationship between STAT3 protein level and thyroid cancer.The quality of the literature was scored by NOS.The literature was selected according to inclusion and exclusion criteria and the data were extracted.Stata 12.0 software was used for Statistical analysis.The heterogeneity test was performed by I2 test.Results:Nine studies with a total of 711 samples were included.Compared with the control group,STAT3 protein level in thyroid cancer patients increased significantly,and the difference was statistically significant(RR=3.366,95%CI:2.657~4.264,P<0.05).Analysis of sample groups in thyroid carcinoma group,the expression of STAT3 in thyroid cancer with lymph node metastasis was higher than that in thyroid cancer without lymph node metastasis(RR=1.313,95%CI:1.149~1.500,P<0.05),and the expression of STAT3 in TNF stage III-IV was higher than that in stage I-II(RR=0.701,95%CI:0.558~0.882,P<0.05).Conclusion:STAT3 protein is highly expressed in thyroid cancer tissues and plays an important role in the occurrence and development of thyroid cancer.STAT3 is expected to become a new therapeutic target for thyroid cancer.
