摘　　要：目的特异性下调肾癌细胞内热休克蛋白70(HSP70)的表达,观察其对肾癌细胞生物学特性的影响,初步探讨其可能机制,为肾癌的治疗探寻新的途径。方法使用针对HSP70的mRNA序列设计的小干扰RNA转染肾癌细胞Caki-1,观察转染后HSP70的mRNA及蛋白表达水平的变化;采用流式细胞术和Transwell小室检测HSP70下调后对肾癌细胞凋亡和迁移能力的影响,检查细胞凋亡和迁移相关蛋白caspase-9剪切体、Bcl-2、Bax和Vimentin的表达变化。结果使用小干扰RNA转染后,Caki-1细胞HSP70的mRNA和蛋白水平均明显下调;特异性下调HSP70表达的实验组Caki-1细胞的凋亡率[(13.21±0.33)%]和迁移抑制率[(43.86±3.02)%]较阴性对照组[分别为(8.85±0.69)%、(4.04±0.72)%)]明显上调(P<0.001);实验组细胞的Vimentin和Bcl-2表达下调,而caspase-9剪切体和Bax表达上调。结论特异性下调HSP70在肾癌细胞内的表达可诱导肾癌细胞凋亡并抑制其迁移能力,HSP70具有作为肾癌治疗靶点的重要研究价值。Objective To specifically down-regulate the expression of heat shock protein 70kD(HSP70)in renal cancer cells,observe its effect on biological characteristics of renal cancer cells,and preliminarily explore its potential mechanism.Methods RNA interference was used to down regulate HSP70 expression in renal cancer cell Caki-1.The changes in mRNA and protein expression of HSP70 were observed after transfection.Subsequently,the effects of HSP70 down-regulation on apoptosis and migration of renal cancer cells were detected.The expressions of apoptosis and migration-related proteins caspase-9 cleaved,Bcl-2,Bax and Vimentin were detected.Results After transfection,the mRNA and protein expression of HSP70 in Caki-1 cells were significantly down regulated.Compared with the negative control group((8.85±0.69)%,(4.04±0.72)%),the apoptosis rate and migration inhibition rate of Caki-1 cells in the experimental group ((13.21±0.33)%,(43.86±3.02)%)increased significantly(P<0.001).Preliminary mechanism study showed that the expressions of Vimentin and Bcl-2 was down-regulated,while the expressions of caspase-9 cleaved and Bax was up-regulated in the experimental group.Conclusions Specific down-regulation of HSP70 expression in renal cancer cells can induce apoptosis and inhibit migration in renal cancer cells.It has important research value as a therapeutic target of renal carcinoma.
