摘　　要：目的:探讨阿尔茨海默病患者外周血清中microRNA-146a、Aβ1-42蛋白、tau蛋白的表达及临床意义。方法:选取我院收治的阿尔茨海默病患者98例作为试验组,另选取50例健康者作为对照组。采用RT-PCR技术检测两组患者外周血清中microRNA-146a相对表达水平,采用双抗体夹心酶联免疫吸附法(ELISA)检测两组患者外周血清中Aβ1-42蛋白及tau蛋白浓度。结果:试验组外周血清中microRNA-146a相对表达量、Aβ1-42蛋白、tau蛋白浓度分别为(2.45±0.43)pg/ml、(37.43±6.75)pg/ml、(22.87±4.51)pg/ml,对照组外周血清中microRNA-146a相对表达量、Aβ1-42蛋白、tau蛋白浓度(0.98±0.12)pg/ml、(51.26±9.24)pg/ml、(13.64±2.91)pg/ml,差异均有统计学意义(P<0.05);试验组患者microRNA-146a与Aβ1-42蛋白呈负相关性(r=-0.882,P<0.05),与tau蛋白无相关性(P>0.05)。结论:阿尔茨海默病患者外周血清中存在microRNA-146a、Aβ1-42蛋白、tau蛋白的差异表达,且microRNA-146a可能是通过调控Aβ1-42蛋白而不是tau蛋白进而参与阿尔茨海默病的发病。Objective:To investigate the expression and clinical significance of microRNA-146a,Aβ1-42 protein and tau protein in peripheral blood of patients with Alzheimer′s disease.Methods:98 patients with Alzheimer′s disease admitted to our hospital were selected as the experimental group(Exp group),and 50 healthy subjects were selected as the control group.RT-PCR was used to detect the relative expression of microRNA-146a in peripheral blood of the two groups.The concentration of Aβ1-42 protein and tau protein in peripheral blood of the two groups were detected by double antibody sandwich enzyme-linked immunosorbent assay(ELISA).Results:The relative expression of microRNA-146a,Aβ1-42 protein and tau protein in the peripheral blood of the Exp group were(2.45±0.43)pg/ml,(37.43±6.75)pg/ml,(22.87±4.51)pg/ml,respectively.The relative expression of microRNA-146a in the peripheral blood of the control group,Aβ1-42 protein,tau protein concentration(0.98±0.12)pg/ml,(51.26±9.24)pg/ml,(13.64±2.91)pg/ml,the difference was statistically significant.(P<0.05).microRNA-146a was negatively correlated with Aβ1-42 protein in the Exp group(r=-0.882,P<0.05),and had no correlation with tau protein(P>0.05).Conclusion:microRNA-146a,Aβ1-42 protein and tau protein are differentially expressed in peripheral blood of patients with Alzheimer′s disease,and microRNA-146a may regulate Aβ1-42 protein in the pathogenesis of Alzheimer′s disease instead of tau protein.
