摘 要:目的探讨粪菌移植对溃疡性结肠炎(UC)小鼠的抗炎作用及可能机制。方法将45只雌性C57BL/6小鼠随机分为对照组、模型组、干预组各15只。模型组及干预组制备UC小鼠模型。造模后第2天开始对干预组进行粪菌移植,其他两组给予等体积生理盐水灌肠,共7 d。采用疾病活动指数(DAI)对小鼠的一般情况进行评价。干预第8天处死小鼠,取结肠组织进行结肠黏膜大体形态损伤指数(CMDI)、组织学评分,并应用流式细胞术检测脾脏组织滤泡辅助性T细胞(Tfh)及滤泡调节性T细胞(Tfr)水平。结果对照组、干预组、模型组的DAI、CMDI、组织学评分依次升高(均P<0.05)。与对照组比较,模型组Tfh及Tfr水平、Tfh/Tfr比值均升高,干预组Tfh及Tfr水平升高,而Tfh/Tfr比值降低(均P<0.05);与模型组比较,干预组Tfr水平升高,而Tfh水平及Tfh/Tfr比值降低(均P<0.05)。结论粪菌移植可减轻UC小鼠肠道黏膜炎症,这可能与其降低促炎症因子Tfh的表达并促进保护因子Tfr的表达有关。Objective To investigate the anti-inflammatory effect and potential mechanism of fecal microbiota transplantation(FMT) in ulcerative colitis(UC) mice. Methods Forty-five female C57 BL/6 mice were randomly divided into control group, model group and intervention group, with 15 mice in each group. UC mouse model was established in the model group and intervention group. From the second day after modeling, FMT was conducted in the intervention group for seven days, and enema with normal saline was performed in the remaining two groups for seven days. Disease activity index(DAI) was employed to evaluate the general condition of mice. On the 8 th day after modeling, the mice were killed, then the colon tissues were removed for assessing colon mucosal damage index(CMDI) and histological score, and the levels of follicular helper T cell(Tfh) and follicular regulatory T cell(Tfr) in spleen tissues were determined by using flow cytometry. Results DAI, CMDI or histological score increased in the control group, the intervention group and the model group successively(all P<0.05). Compared with the control group, the model group exhibited higher Tfh and Tfr levels with a higher Tfh/Tfr ratio, whereas the intervention group possessed higher Tfh and Tfr levels with a lower Tfh/Tfr ratio(all P<0.05);compared with the model group, the intervention group yielded a higher Tfr level, as well as lower Tfh level and Tfh/Tfr ratio(all P<0.05). Conclusion FMT can alleviate the inflammation of colon mucosa in UC mice, which may be related to reducing proinflammatory factor Tfh expression and promoting protective factor Tfr expression.
