摘 要:目的:研究龙蛭汤对大鼠局灶性脑缺血再灌注损伤后脑组织的保护作用及其对内质网应激(ERS)标志蛋白葡萄糖调节蛋白78(GRP78)和特异性蛋白C增强子结合蛋白同源蛋白(CHOP)mRNA表达的影响。方法:将48只大鼠随机分为4组,假手术组、模型组、丁苯酞组及龙蛭汤组,每组12只。除假手术组以外,其余3组均采用改良Longa线栓法建立右侧脑缺血再灌注损伤大鼠模型。造模成功后丁苯酞组、龙蛭汤组大鼠分别给予丁苯酞(40 mg/kg·d^-1)、龙蛭汤(13.6 g/kg·d^-1)灌胃,假手术组和模型组用等量生理盐水灌胃,连续灌胃3 d。术后第3天对各组大鼠进行神经功能缺损评分,并采用2,3,5-氯化三苯四氮唑(TTC)染色法测定脑梗死体积,苏木精-伊红(HE)染色法观察海马区脑组织病理形态变化,实时荧光定量PCR(qPCR)法比较各组GRP78和CHOP mRNA表达。结果:与假手术组相比,模型组可见明显神经功能缺损及大面积脑梗死区域,海马区神经细胞损伤严重,海马组织中GRP78和CHOP mRNA相对表达量明显升高(P<0.05);与模型组相比,龙蛭汤组和丁苯酞组的神经功能缺损评分、脑梗死体积百分比、海马组织中GRP78和CHOP mRNA相对表达量均明显降低(P<0.05),海马区神经细胞形态明显改善。结论:龙蛭汤可减轻大鼠脑缺血再灌注损伤,其机制可能与下调脑组织中GRP78和CHOP基因的表达有关。Objective:To investigate the protective effect of Longzhi decoction on brain tissues after focal cerebral ischemia-reperfusion injury in rats and its effect on the mRNA expression of glucose-regulated protein 78(GRP78)and C enhancer binding protein homologous protein(CHOP).Methods Forty-eight rats were randomly divided into 4 groups:sham operation group,model group,butylphthalide group,and Longzhi decoction group,with 12 rats in each group.Except for the sham operation group,the other three groups were all given the modified longa thread occlusion method to establish the right cerebral ischemia-reperfusion injury model.After successful modeling,the rats in the butylphthalide group and the Longzhi decoction group were given butylphthalide(40 mg/kg·d^-1)and longzhi decoction(13.6 g/kg·d^-1)by intragastric administration,respectively,the sham operation group and the model group were given with normal saline by intragastric administration,drugs was administered once a day for 3 consecutive days.At the 3 rd day after treatment,the neurologic deficit score was evaluated.2,3,5-triphenyltetrazolium chloride(TTC)staining was used to determine the cerebral infarction volume.HE staining was used to observe the pathological changes of brain tissues in hippocampus.Real-time quantitative PCR(qPCR)was employed to compare the expression of GRP78 and CHOP mRNA in each group.Results:Compared with the sham operation group,the model group showed obvious neurological deficits,large areas of cerebral infarction,severe nerve cells damaged,and the significant increased relative expressions of GRP78 and CHOP mRNA in hippocampal tissues(P<0.05).Compared with the model group,the neurological deficit scores,volume percentage of cerebral infarction,and relative expression of GRP78 and CHOP mRNA in hippocampal tissues were significantly reduced(P<0.05),while the neuronal morphology of hippocampus was significantly improved in the Longzhi decoction group and butylphthalide group.Conclusion:Longzhi decoction can alleviate cerebral ischemia-re
