摘  要:目的基于RNA干扰技术分析微小染色体维持蛋白7(MCM7)在涎腺腺样囊性癌(SACC)中的作用.方法应用Western-blot检测SACC-83、SACC-LM中MCM7蛋白表达,再将SACC-LM分别转染3个序列的siRNA-MCM7和siRNA-NegativeControl,利用Western-blot验证转染效率,选择沉默效率较高的2组进行实验.细胞克隆、CCK-8实验测定细胞增殖情况,Transwell小室验证各组细胞侵袭情况,细胞划痕实验验证细胞迁移情况.结果SACC-LM中MCM7蛋白表达高于其在SACC-83中的表达,干扰组细胞中MCM7蛋白水平低于对照组,与对照组比较,干扰组细胞增殖能力更低、细胞侵袭和迁移数目更少.结论沉默MCM7表达可抑制SACC-LM细胞的增殖、侵袭及迁移能力,MCM7可能作为癌基因促进SACC的进展.Objective To investigate the role of MCM7 in salivary glands(SACC)adenoid cystic carcinoma based on RNA interference.Methods The expression of MCM7 protein in SACC-83 and SACC-LM was detected by Western-blot.After transfected with three sequences of siRNA-MCM7 and siRNA-Negative Control,the protein level of MCM7 was detected by Western blot,using Western-blot to verify transfection efficiency,and two groups with higher silencing efFiciency for experiments were selected.Cell cloning,CCK-8 assay were used to determine cell proliferation,Transwell chamber was used to verify the invasion of each group,and cell scratch assay was used to verify cell migration.Results The expression ofMCM7 protein in SACC-LM was higher than that in SACC-83.The level of MCM7 protein in the interference group was lower than that in the control group.Compared with the control group,the interference group had lower cell proliferation ability and fewer cell invasion and migration.Conclusion Silencing MCM7 expression can inhibit the proliferation,invasion and migration ofSACC-LM cells,and MCM7 may act as an oncogene to promote the progression of SACC.
