摘  要:目的研究新疆芹菜根提取物(AGRE)对高脂血症(HLP)大鼠血脂水平和抗氧化能力的影响。方法采用灌服高脂乳剂建立SD大鼠HLP模型,将40只HLP大鼠随机分为模型组(高脂乳剂10 mL/kg体质量),阳性对照组(血脂康0.14 g/kg体质量+高脂乳剂),AGRE高、中、低剂量组(AGRE0.09 g/kg体质量+高脂乳剂、AGRE0.18 g/kg体质量+高脂乳剂、AGRE0.27 g/kg体质量+高脂乳剂),连续给药6周;另取8只正常SD大鼠设为空白对照组(生理盐水,10mL/kg),连续给予6周;AGRE预防组于造模前给予AGRE(0.54 g/kg)4周,造模期间继续给予AGRE 3周。实验结束后各组大鼠经腹主动脉采血,检测血清中总胆固醇(TC)、甘油三脂(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平,测定肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)含量,并用光学显微镜观察各组大鼠肝脏组织病理学变化。结果与模型组比较,AGRE各剂量组大鼠血清中TC、TG水平降低,HDL-C水平升高,差异有统计学意义(P<0.01),SOD含量升高,差异有统计学意义(P<0.05);AGRE预防组大鼠血清中TC、TG水平降低,SOD含量升高,差异有统计学意义(P<0.05)。AGRE预防组和AGRE各剂量组大鼠肝小叶结构明显,肝细胞排列较整齐,肝细胞脂肪变性范围小且程度较轻。结论AGRE具有改善HLP大鼠血脂水平,增强抗氧化作用的效果,具有调节血脂的活性。Objective To study the hypolipidemic effect of the extract of Xinjiang Apium graveolens L.root(AGRE)in hyperlipidemia rats and its antioxidant activity.MethodsThe hyperlipidemia model of rat was established by high-fat dairy feeding for 3 weeks.40 hyperlipidemi rats were randomly allocated into model group(high-fat emulsion,10 mL/kg),positive control group(Xuezhikang,10 mL/kg)and AGRE low(0.09 g/kg),middle(0.18 g/kg)and high dose(0.27 g/kg)dose groups.The rats in the intervention groups were continuously administered drugs and high-fat emulsion for 6 weeks,and the high-fat diet(HFD)group was continuously received daily gavage of 10 mL/kg high-fat emulsion for 6 weeks only.The prevention group was continuously administered AGRE(0.5 g/kg)for 4 weeks before the high-fat model establishment and for another 3 weeks during the establishment.After rats were sacrificed,the serum levels of total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C)and the levels of superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-Px)in the liver tissue homogenates were measured.The pathological examination of liver tissues were performed.ResultsCompared with the model group,the levels of TC and TG reduced significantly(P<0.01)and the levels of HDL-C and SOD increased significantly(P<0.05)in AGRE groups.The levels of TC and TG reduced significantly(P<0.05)and the levels of SOD increased significantly(P<0.05)in the prevention group.The rat liver of the AGRE groups and prevention group had a distinct lobular structure,neatly arranged hepatocytes,paralleled by lesser range and the milder fatty degeneration of liver tissues.ConclusionThere is a significant lipid-modulation and enhance antioxidant activity in hyperlipidemia rats by prophylactic and therapeutic administration.
