摘  要:目的观察人肌源性干细胞(MDSCs)的生物特性及其治疗假肥大肌营养不良症模型鼠(mdx)的再生潜能。方法预贴壁法分离细胞,采用免疫荧光,Real-time PCR和Western Blotting法检测相关成肌因子;并观察其移植治疗mdx后的dystrophin蛋白的变化。结果与D1-hMDCs相比,D6-hMDCs和D8-hMDCs的CD56、CD45和CD184均呈低表达;desmin和MHC呈显著高表达;移植到mdx鼠后,均能增强dystrophin蛋白表达,但D6-hMDCs与D8-hMDCs间没有差异。结论 D6-hMDCs和D8-hMDCs具有相似的生物学特性,属MDSCs,均比D1-hMDCs具有更强的成肌分化能力和再生潜能。Objective To identify the biological characteristics and the regenerative potential of human muscle-derived stem cells(hMDSCs)in the treatment of mdx mice. Methods The expression of myogenic markers were detected via immunofluorescence(IF),Real-time PCR,and Western Blotting. After cell transplantation,dystrophin expression was observed. Results Comparing to D1-hMDCs,D6-hMDCs and D8-hMDCs showed higher expression of CD56,CD45,CD184,desmin,MHC and dystrophin,while there was no difference between D6-hMDCs and D8-hMDCs. Conclusion D6-hMDCs and D8-hMDCs show similar biological characteristics and belong to MDSCs. They have stronger ability of myogenic differentiation and regeneration than D1-hMDCs.
