摘 要:目的:观察牙周炎基因疫苗pVAX1-HA2-FimA、pVAX1-HA2-FimA/IL-15对SD大鼠实验性牙周炎的保护作用。方法:54只雄性SD大鼠随机分为9组,对照组:生理盐水组(A组)、空载质粒pVAX1组(B组)、牙周病组(C组);实验组:50μg、100μg、150μg pVAX1-HA2-FimA组(D、E、F组)和50μg、100μg、150μg pVAX1-HA2-FimA/IL-15组(G、H、I组)。鼻滴免疫SD大鼠后建立实验性牙周炎模型,采用RT-qPCR法检测牙龈中炎症因子肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、基质金属蛋白酶8(matrix metalloproteinase-8,MMP-8)m RNA相对表达量;立体显微镜观察并测量牙槽骨吸收量。结果:①各实验组牙龈组织中TNF-α、MMP-8 mRNA低于牙周病组,且差异有统计学意义(P<0.05);②相同剂量的pVAX1-HA2-FimA/IL-15组较pVAX1-HA2-FimA组大鼠牙槽骨吸收量少,且差异有统计学意义(P<0.05)。结论:牙周炎基因疫苗pVAX1-HA2-FimA和pVAX1-HA2-FimA/IL-15对SD大鼠实验性牙周炎具有保护作用。Objective:This study aims to observe the protective effect of periodontitis gene vaccine pVAX1-HA2-FimA and pVAX1-HA2-FimA/IL-15 in SD rats with experimental periodontitis. Methods:Fifty-four male SD rats were randomly divided into 9 groups:Control group(normal saline group,group A),empty plasmid p VAX1 group(group B),periodontal disease group(group C),Experimental group[50 μg,100 μg,150 μg pVAX1-HA2-FimA group(D,E,F group)and 50 μg,100 μg,150 μg pVAX1-HA2-FimA/IL-15 group(G,H,I group)]. Experimental periodontitis model was established after nasal drip immunization of SD rats,and the relative expression levels of TNF-α and MMP-8 mRNA in gingiva were detected by RT-qPCR. The absorption of alveolar bone was observed and measured by stereomicroscopy. Results:①The TNF-α and MMP-8 mRNA in the gingival tissues of each experimental group were lower than that in the periodontal disease group,and the differences were statistically significant(P < 0.05). ②Compared with the pVAX1-HA2-FimA group,the amount of alveolar bone absorption in the pVAX1-HA2-FimA/IL-15 group with the same vaccine dose was lower,and the differences were statistically significant(P < 0.05). Conclusion:The periodontitis gene vaccines pVAX1-HA2-FimA/IL-15 and pVAX1-HA2-FimA have protective effects on SD rats with experimental periodontitis.
