摘 要:目的探讨miR-126与靶基因GOLPH3在胃癌中的表达及潜在机制。方法选取45例胃癌患者和43例健康体检者为研究对象,qRT-PCR法检测患者和健康组血液中miR-126、GOLPH3、mTOR表达量;免疫组化法检测胃癌组织及癌旁正常胃组织GOLPH3、mTOR表达;分析检测指标与胃癌临床病理特征的关系。生物信息软件预测GOLPH3是miR-126潜在靶基因,利用双荧光素酶报告实验鉴定靶向关系。胃癌SGC7901细胞分组:转染miR-126过表达质粒(miR-126过表达组)、空质粒(空载组)、未转染(空白组),并检测miR-126过表达组GOLPH3表达量。结果癌患者血液中miR-126低于健康组,而GOLPH3、mTOR则高于健康组(P<0.05);癌组织中GOLPH3、mTOR表达均高于癌旁正常胃组织(P<0.05);所测指标与临床TNM分期、淋巴结转移均相关(P<0.05)。双荧光素酶报告实验提示GOLPH3是miR-126靶基因。miR-126过表达组GOLPH3表达量较空载组明显降低(P<0.01)。结论miR-126在胃癌中作为抑癌因子,部分是通过靶向调节GOLPH3发挥作用,并且可能介导mTOR相关通路参与肿瘤的进展。Objective To investigate the expressions and potential mechanism of miR-126 and target gene GOLPH3 in gastric cancer.Methods A total of 45 patients with gastric cancer and 43 healthy people were selected as subjects.To compare the expressions of miR-126,GOLPH3 and mTOR in their blood were detected by qRTPCR.The expressions of GOLPH3 and mTOR in cancer tissues and adjacent normal gastric tissues were examined by immunohistochemistry.The relationship between the levels of the indicators and clinicopathological features was analyzed.GOLPH3 was predicted to be a potential target gene of miR-126 by RNAHybrid and the relationship was identified by dual-Luciferase Reporter assay.Human gastric cancer cells(SGC7901)were divided into miR-126 increase group(transfection group),blank vector group,blank control group,the expression of GOLPH3 was detected in miR-126 increase group.Results The expression of miR-126 in the blood of patients was lower than that of healthy group,GOLPH3 and mTOR were higher than those in the healthy group(P<0.05).The positive expression rates of GOLPH3 and mTOR in cancer tissue were significantly increased compared with those in the normal gastric tissue adjacent to cancer(P<0.05).The indexes were significantly related with clinical TNM stage and lymph node metastasis in patients with gastric cancer(P<0.05).The dual-Luciferase Report experiment suggested GOLPH3 was the target gene of miR-126.The expression level of GOLPH3 in the miR-126 increase group was significantly lower than that in the blank vector group(P≤0.01).Conclusion As an anti-oncogene,miR-126 suppressed gastric cancer in part by targeting GOLPH3 via activated downstream mTOR signaling.
