摘  要:研究miRNA-143通过调节PTEN/Akt途径对鼻咽癌HONE1细胞生物学特征及紫杉醇敏感性的影响。该研究实验细胞由上海北诺生物科技有限公司提供的HONE1细胞株,3份HONE1细胞株分别为miRNA-143转染组,miRNA-143抑制剂转染组以及空白组。比较不同组别的miRNA-143含量、不同组别HONE1细胞的PTEN、Akt水平以及不同组别HONE1细胞不同转染时间的凋亡情况。miRNA-143转染组,miRNA-143抑制剂转染组以及空白组HONE1细胞的miRNA-143含量分别为(9.23±3.90)、(4.09±1.12)以及(10.43±2.19),3组患者的miRNA-143含量之间的差异存在统计学意义(F=10.163,P=0.000),通过两两比较,miRNA-143转染组和空白组miRNA-143含量显著高于miRNA-143抑制组,且miRNA-143转染组和空白组miRNA-143含量之间的差异不存在统计学意义(P>0.05),3组患者的PTEN、Akt水平差异存在统计学意义(P<0.05),通过两两比较,miRNA-143抑制剂转染组患者的PTEN水平显著高于miRNA-143转染组以及空白组,Akt水平显著低于miRNA-143转染组以及空白组,miRNA-143抑制剂转染组不同转染时间的HONE1细胞增殖情况显著低于miRNA-143转染组以及空白组,差异存在统计学意义(P<0.05)。通过对凋亡率的比较,miRNA-143抑制剂转染组不同转染时间的HONE1细胞凋亡情况显著高于miRNA-143转染组以及空白组。miRNA-143可通过对PTEN/Akt信号通路的抑制性作用,有效降低肿瘤细胞的增殖以及迁移,在对鼻咽癌的紫杉醇治疗中,可将miRNA-143作为治疗的靶点,评估鼻咽癌的紫杉醇治疗的敏感浓度。To study the effects of microRNA-143 on the biological characteristics and taxol sensitivity of nasopharyngeal carcinoma HONE1 cells by regulating PTEN/Akt pathway,three HONE1 cell lines were obtained from the HONE1 cell lines provided by Shanghai Beino Biotechnology Co.,Ltd.and they were microRNA-143 transfection group,microRNA-143 inhibitor transfection group and blank group respectively.The content of microRNA-143 in different groups,the levels of PTEN and Akt in HONE1 cells of different groups and the apoptosis of HONE1 cells of different groups at different transfection time were compared.The contents of microRNA-143 in HONE1 cells of the three groups were(9.23±3.90),(4.09±1.12)and(10.43±2.19),respectively.There were significant differences in the contents of microRNA-143 among the three groups(F=10.163,P=0.000).By comparing the two groups,microRNA-143 was transfected into HONE1 cells.The content of microRNA-143 in group A and blank group was significantly higher than that in group A,and there was no significant difference in the content of microRNA-143 between transfection group and blank group(P>0.05).The levels of P TEN and Akt in three groups were significantly different(P<0.05).By comparing two groups,the content of microRNA-143 inhibitor transfection group was significantly higher than that in control group(P<0.05).PTEN level was significantly higher than that of microRNA-143 transfection group and blank group.Akt level was significantly lower than that of microRNA-143 transfection group and blank group.The proliferation of HONE1 cells in microRNA-143 inhibitor transfection group was significantly lower than that of microRNA-143 transfection group and blank group at different transfection time(P<0.05).By comparing the apoptotic rate,the apoptotic rate of HONE1 cells transfected with the inhibitor of microRNA-143 at different time was significantly higher than that of the microRNA-143 transfection group and the blank group.MiRNA-143 can effectively reduce the proliferation and migration of tumor cell
