摘 要:目的观察豆豉对雄激素源性脱发(AGA)小鼠血液内源性代谢物的干扰规律,从代谢组学方面探讨豆豉防治脱发的作用机制。方法除空白组外,其余各组均在小鼠背部平行于脊柱区皮下注射丙酸睾酮溶液建立AGA小鼠模型,阳性药组灌胃非那雄胺,给药组灌胃高、中、低剂量的豆豉溶液,空白组和模型组灌胃蒸馏水。给药周期为10周,给药期间观察小鼠的状态。给药治疗后,收集小鼠血清,应用UPLC/Q-TOF-MS技术结合多元统计学分析方法,阐释其代谢谱的变化规律。结果豆豉可以明显改善AGA小鼠的脱发现象,在正负离子的模式下,共筛选出12个差异性标志物,包括全反式维甲酸、9-顺式-视黄醛、鞘氨醇-1-磷酸、亚油酸、磷脂酰胆碱等。结论豆豉改善AGA脱发小鼠的机制可能与甘油磷脂代谢、鞘脂代谢、亚油酸代谢、视黄醇代谢、氨基酸代谢等代谢路径有关。Objective To observe the interference of soybean meal on endogenous metabolites in the blood of mice with androgenetic alopecia(AGA), and discuss the mechanism of action of soybean meal on prevention and treatment of hair loss from metabolomics. Methods Except for the blank group, all other groups were injected testosterone propionate solution on the back of mice, parallel to the spinal region to establish AGA mouse models. The positive drug group were administered finasteride, the high-, medium-, and low-dose solutions of cardamom were given to the drug group, and distilled water to the blank group and the model group, for 10 weeks. The state of the mice was observed during the administration. The serum of the mice was collected, and UPLC/Q-TOFMS technique was combined with multivariate statistical analysis to explain the changes in metabolic profiles. Results Soybean meal significantly improved the hair loss in AGA mice. In the positive and negative ion mode, 12 differential markers were screened, including all-trans retinoic acid, 9-cis-retinal, sphingosine-1-phosphate, linoleic acid, and phosphatidylcholine. Conclusion The mechanism of soybean meal improving hair loss in AGA mice may be related to the metabolic pathways, such as glycerophospholipid metabolism, sphingolipid metabolism, linoleic acid metabolism, retinol metabolism, and amino acid metabolism.
