摘 要:为减少抗逆分子育种中因引入外源抗性基因造成的基因多效性,降低外源基因对植物的伤害,本研究设计并合成逆境响应顺式元件不同组合的启动子,将其连上GUS报告基因后转化为拟南芥野生型Col-0;通过转基因拟南芥中GUS报告基因的表达强度分析各种启动子对干旱和盐害的响应情况,筛选出了在正常条件下无渗漏、在干旱和盐害胁迫下被诱导的启动子AXpro。然后,利用发根农杆菌介导转化大豆,发现干旱诱导生物钟基因GmTIC的沉默能提高大豆的抗旱能力。研究结果为农林业的抗逆分子育种提供了理论基础和应用方法。In order to reduce the gene practicability that caused by the introduction of exogenous resistance genes in the breeding of stress-resistant molecules,and the damage of foreign genes to plants,we designed and cloned a set of promoters consisting of various combination of stress-responding cis-elements.Then,they were marked with the GUS reporter gene and the resulting binary vectors were introduced into Arabidopsis wild type Col-0.GUS staining of transgenic Arabidopsis showed that different practicability promoters responded to drought and salt stresses at different extents,with an exception of AXpro,which did not display any detectable activities in normal growth conditions,but was induced in drought and salt stress conditions,suggesting that it was a stress-inducible promoter.We further investigated the application of these promoters in soybean by exploiting hairy root transformation mediated by Agrobacterium rhizogenes.We found that drought-induced RNAi-silencing of clock gene GmTIC(Glycine max Time for Coffee)could increase drought resistance in soybean.The results provide practicability basis and application method for stress-resistant molecular breeding in agroforestry.
