摘  要:目的探讨沉默周期蛋白依赖性激酶6(CDK6)基因对人卵巢癌HO-8910细胞增殖和粘附能力的影响。方法设计合成5条特异性CDK6 shRNA转染HO-8910细胞,检测转染前后细胞内CDK6 mRNA的表达变化;选出抑制作用最强的CDK6-shRNA转染HO-8910细胞,用MTT法检测转染前后HO-8910细胞增殖和粘附能力的变化。结果CDK6 shRNA-608表达载体明显抑制了HO-8910细胞中CDK6基因mRNA的表达,转染48 h后,细胞的增殖能力和粘附能力明显下降,与无关序列组和空白对照组比,差异显著(P<0.05)。结论沉默CDK6基因可抑制卵巢癌HO-8910细胞的增殖和粘附能力。Objective To investigate the effect of CDK6 gene silencing on the proliferation and adhesion of human ovarian cancer HO-8910 cells.Methods Five specific CDK6 shRNAs were designed and synthesized to transfect HO-8910 cells.The expression of CDK6 mRNA in HO-8910 cells before and after transfection was detected.The most inhibitory CDK6-shRNA was selected to transfect HO-8910 cells.The proliferation and adhesion of HO-8910 cells before and after transfection were detected by MTT method.Results CDK6 shRNA-608 expression vector significantly inhibited the expression of CDK6 gene mRNA in HO-8910 cells.48 hours after transfection,the cell proliferation and adhesion ability decreased significantly,compared with the unrelated sequence group and the blank control group,the difference was significant(P<0.05).Conclusion CDK6 gene silencing can inhibit the proliferation and adhesion of HO-8910 cells.
