摘 要:为了鉴定和分析五指山猪背最长肌和半腱肌中差异表达的lncRNA和mRNA,试验以6月龄五指山猪阉割公猪为试验动物,在无菌状态下逐头采集背最长肌和半腱肌样品,采用RNA-Seq文库和生物信息学方法对五指山猪肌肉组织中的lncRNA和mRNA进行分析筛选,运用DESeq软件鉴定背最长肌和半腱肌中差异表达的lncRNA和mRNA,并对差异表达的lncRNA进行靶基因预测分析。结果表明:在五指山猪肌肉组织中共鉴定出4086个lncRNA,其中正义型2203个、反义型197个、内含子型130个、基因间型1556个。对比测序获得的lncRNA和mRNA发现,lncRNA和mRNA的长度都主要分布在600~1200 nt之间和3000 nt以上。在背最长肌与半腱肌间,存在82个差异表达的mRNA,其中前10位分别为ZIC4、ZIC1、PITX1、SIM1、PVALB、SEC14L5、SERPINB11、PRXL2B、ZNF862、MYBPH;存在32个差异表达的lncRNA,其中2个仅在背最长肌中表达,1个仅在半腱肌中表达,29个在两种肌肉组织中共表达;获得差异表达的lncRNA靶基因共70个,包括顺式作用的靶基因和反式作用的靶基因。说明五指山猪背最长肌和半腱肌具有不同表达水平的lncRNA和mRNA,两种肌肉组织中均存在特异性表达的lncRNA。The objective of this study was to identify and analyze the differentially expressed lncRNAs and mRNAs between longissimus dorsi and semitendinosus in Wuzhishan pig.In this study,6 months old castrated Wuzhishan male pigs were selected as experimental animals.The longissimus dorsi and semitendinosus samples were collected under sterile condition,respectively.RNA-Seq and bioinformatics assays were used to analyze and filter lncRNAs and mRNAs in skeletal muscle tissue of Wuzhishan pig.The differentially expressed lncRNAs and mRNAs between longissimus dorsi and semitendinosus were identified by DESeq.The targe genes of differentially expressed lncRNAs were predicted and analyzed.The results showed that 4086 lncRNAs were identified in skeletal muscle tissue of Wuzhishan pig.Among them,2203 were sense lncRNAs,197 were antisense lncRNAs,130 were intronic lncRNAs,and 1556 were intergenic lncRNAs.LncRNA and mRNA were mainly distributed in the range of 600-1200 nt and≥3000 nt.A total of 82 mRNAs were differentially expressed between longissimus dorsi and semitendinosus.The top 10 differentially expressed mRNAs were ZIC4(Zic family member 4),ZIC1(Zic family member 1),PITX1(paired like homeodomain 1),SIM1(SIM bHLH transcription factor 1),PVALB(parvalbumin),SEC14L5(SEC14 like lipid binding 5),SERPINB11(serpin family B member 11),PRXL2B(peroxiredoxin like 2B),ZNF862(zinc finger protein 862)and MYBPH(myosin binding protein H),respectively.A total of 32 differentially expressed lncRNAs were identified.Among them,2 lncRNAs were only expressed in longissimus dorsi,and 1 lncRNAs were only expressed in semitendinosus,and 29 lncRNAs were co-expressed in two muscle tissues.There were 70 target genes of differentially expressed lncRNAs,including cis-acting target genes and trans-acting target genes.This study proved that LncRNAs and mRNAs expression levels were significantly different between longissimus dorsi and semitendinosus in Wuzhishan pig.
