摘　　要：目的:研究3'-大豆苷元磺酸钠对大鼠脑缺血再灌注损伤后脑组织中基质金属蛋白酶-3、基质金属蛋白酶-9、紧密连接蛋白ZO-1及紧密连接蛋白OCLN mRNA表达的影响。方法:建立大鼠大脑中动脉栓塞再灌注模型,取缺血再灌注后大鼠缺血半损伤区皮层脑组织,实时定量PCR方法检测组织中MMP-3、MMP-9、ZO-1及OCLN mRNA的表达。结果:与模型组相比,3'-大豆苷元磺酸钠治疗能显著降低MMP-3、MMP-9 mRNA的表达水平,能显著提高ZO-1、OCLN mRNA的表达水平。结论:3'-大豆苷元磺酸钠可能通过抑制MMP-3、MMP-9 mRNA的表达和促进ZO-1、OCLN mRNA的表达,从而减轻大鼠脑缺血再灌注后的损伤。Objective:To investigate the effects of 3'-daidzein sulfonate sodium(DSS)on the mRNA expression of matrix metalloproteinase-3,matrix metalloproteinase-9,ZO-1 and OCLN after Cerebral ischemia-reperfusion injury in rats.Methods:The rat cerebral ischemia-reperfusion injury mode was established by middle cerebral artery occlusion,remove the cortex of the ischemic semi injured area after 120 minutes of ischemia and 24 hours of reperfusion.The mRNA expression levels of MMP-3,MMP-9,ZO-1 and OCLN on half injury brain tissue were detected by Real time-PCR.Results:Compared with the model group,DSS significantly reduced the mRNA expression of MMP-3 and MMP-9,and significantly increased the mRNA expression of tight junction protein ZO-1 and occludin.Conclusions:The protective effects of DSS on cerebral ischemia-reperfusion injury in rats might be associated with its potential to reduce the mRNA expression of MMP-3 and MMP-9 and to increase the mRNA expression of ZO-1 and OCLN.
