摘 要:目的观察表皮生长因子(epidermal growth factor,EGF)及其受体(epidermal growth factor receptor,EGFR)经电针干预后在近视豚鼠睫状肌中的表达变化,探讨其在近视发生发展中的作用。方法将90只2周龄健康三色短毛豚鼠随机分为正常对照(normal control,NC)组、透镜诱导型近视(lens-induced myopia,LIM)组和透镜诱导型近视电针(lens-induced myopia+electroacupuncture,LIM+EA)组,每组各30只。NC组豚鼠正常饲养,不做任何干预;LIM组和LIM+EA组豚鼠右眼均戴-6 D透镜诱导近视,左眼作为自身对照不做任何处理;LIM+EA组豚鼠在戴镜同时选取双侧太阳穴、合谷穴进行电针干预。各组豚鼠造模后2周、4周进行屈光度和眼轴长度的检测,分别制备组织病理切片观察睫状肌的形态;通过定量聚合酶链反应(quantitative polymerase chain reaction,q-PCR)和酶联免疫吸附实验(enzyme-linked immunosorbent assay,ELISA)检测各组豚鼠睫状肌中EGF mRNA、EGFR mRNA及其蛋白表达水平。结果造模后2周、4周,LIM组和LIM+EA组造模眼分别与自身对照眼及NC组右眼相比,近视屈光度均增加,眼轴均延长,LIM+EA组造模眼较LIM组造模眼近视屈光度减小,眼轴延长减缓,差异均有统计学意义(均为P<0.05)。病理检测结果显示,NC组睫状肌纤维完整、排列有序;LIM组睫状肌纤维断裂、溶解,排列混乱;LIM+EA组睫状肌纤维相对完整,排列有序。q-PCR及ELISA检测结果表明,LIM组右眼睫状肌中EGF mRNA、EGFR mRNA及其蛋白表达水平与NC组和LIM+EA组相比均明显降低(均为P<0.05),而LIM+EA组和NC组EGF mRNA、EGFR mRNA及其蛋白表达水平差异均无统计学意义(均为P>0.05)。结论电针干预近视豚鼠可影响睫状肌中EGF和EGFR的表达水平并能延缓近视发展。Objective To observe the expression changes of epidermal growth factor(EGF)and epidermal growth factor receptor(EGFR)in ciliary muscle of lens-induced myopic(LIM)guinea pigs after electroacupuncture intervention,and to explore their roles in the development of myopia.Methods Ninety healthy three-colored short-hair guinea pigs(two-week-old)were randomly divided into the normal control(NC)group,LIM group and LIM+electroacupuncture(LIM+EA)groups,30 guinea pigs in each group.Guinea pigs in NC group were normally raised,and did not receive any treatment.While right eyes of guinea pigs in both LIM and LIM+EA groups worn-6D lens to induce myopia,and the fellow eyes treated nothing were as self-control.Guinea pigs in LIM+EA group received electroacupuncture stimulation at bilateral acupoints of Hegu and Taiyang simultaneously.The diopter and axial length of guinea pigs in each group were measured 2 weeks and 4 weeks after modeling.Histopathological section was prepared for observing morphology of ciliary muscles.The expression of EGF and EGFR at mRNA and protein levels in ciliary muscle were detected by quantitative polymerase chain reaction(q-PCR)and enzyme-linked immunosorbent assay(ELISA).Results At 2 weeks and 4 weeks after modeling,the myopia diopter and axial length increased in induction eyes in both LIM and LIM+EA groups;Myopia diopter and axial length decreased in induction eyes in LIM+EA group comparing with LIM group,and the differences were statistically significant(all P<0.05).Pathological analysis revealed that the fibers of ciliary muscle were intact and orderly for guinea pigs in NC group,broken,dissolved and arranged disorderly in LIM group,while relatively intact and orderly in LIM+EA group.q-PCR and ELISA results showed that the expression of EGF and EGFR at mRNA and protein levels in ciliary muscle of the right eyes in LIM group were significantly lower than those in the NC and LIM+EA groups(all P<0.05),whereas there was no significant difference in the expression of EGF and EGFR at mRNA and protein l
