摘  要:目的研究大蒜素通过调节Toll样受体4/髓样分化因子88/核因子-κB(TLR4/My D88/NF-κB)信号通路对颈动脉狭窄大鼠的保护作用。方法将Wistar大鼠160只,随机分为假手术组(n=40)、模型组(n=40)、对照组(n=40)和实验组(n=40)。模型组、对照组及实验组大鼠均用2F Fogarty球囊导管于右侧颈动脉进行球囊拉伤的方法构建颈动脉狭窄大鼠模型,同时假手术组大鼠进行相同的手术操作,但不插入2F Fogarty球囊。造模成功后,实验组大鼠经尾静脉给予大蒜素(18 mg·kg^-1·d^-1),对照组大鼠经尾静脉给予前列地尔(72μg·kg^-1·d^-1),假手术组与模型组注射等量生理盐水,连续干预3周。用酶联免疫吸附(ELISA)法检测血清单核细胞趋化因子-1(MCP-1)、C反应蛋白(CRP)、前列腺素E2(PGE2)及转化生长因子-β(TGF-β)水平;用苏木精-伊红(HE)染色观察大鼠右侧颈动脉病理变化;用蛋白质印迹(WB)法检测各组大鼠右侧颈动脉组织TLR4、MyD88、NF-κB蛋白表达。结果假手术组、模型组、实验组及对照组大鼠血清MCP-1水平分别为(63.47±3.92),(93.65±7.84),(72.34±5.17),(68.83±4.51)ng·L^-1;CRP水平分别为(2.13±0.64),(5.06±1.32),(3.75±1.24),(2.95±1.12)ng·L^-1;PGE2水平分别为(42.36±2.51),(85.22±4.82),(62.16±3.73),(51.06±3.43)ng·L^-1;TGF-β水平分别为(14.32±1.25),(43.87±2.92),(28.33±3.11),(20.62±2.23)ng·L^-1。模型组大鼠血清MCP-1、CRP、PGE2、TGF-β水平及右侧颈动脉组织TLR4、MyD88、NF-κB蛋白相对表达量均显著高于假手术组,且实验组及对照组低于模型组,对照组低于实验组(均P<0.05)。结论大蒜素可降低颈动脉狭窄大鼠炎性因子释放,抑制炎症反应,进而控制颈动脉狭窄进展,其作用机制可能与抑制TLR4/MyD88/NF-κB通路信号转导相关。Objective To investigate the protective effect of allicin on carotid artery stenosis rats by regulating Toll-like receptor 4/myeloid differentiation factor 88/nuclear factor-κB(TLR4/My D88/NF-κB)signaling pathway.Methods A total of 160 Wistar rats were randomly divided into sham-operation group(n=40),model group(n=40),test group(n=40)and control group(n=40).In model group,control group and test group,the rat model of carotid artery stenosis was constructed by 2 F Fogarty balloon catheter in the right carotid artery,and rats in sham-operation group performed the same surgical procedure,but no 2 F Fogarty balloon was inserted.After successful modeling,rats in test group were given allicin 18 mg·kg^-1·d^-1 via the tail vein,the rats in control group were given alprostadil 72μg·kg^-1·d^-1 via the tail vein,sham-operation group and model group were injected with the same amount of normal saline for 3 weeks.Serum monocyte chemoattractant-1(MCP-1),C-reactive protein(CRP),prostaglandin E2(PGE2)and transforming growth factor-β(TGF-β)levels were measured by enzyme-linked immunosorbent assay(ELISA),the pathological changes of the right carotid artery in rats was observed by hematoxylin-eosin(HE)staining,Western blot(WB)method was used to detect the expression of TLR4,MyD88 and NF-κB in the right carotid arteries of each group rats.Results The serum MCP-1 levels in sham-operation group,model group,test group and control group were(63.47±3.92),(93.65±7.84),(72.34±5.17),(68.83±4.51)ng·L-1,the CRP levels were(2.13±0.64),(5.06±1.32),(3.75±1.24),and(2.95±1.12)ng·L-1,the PGE2 levels were(42.36±2.51),(85.22±4.82),(62.16±3.73),(51.06±3.43)ng·L-1,the levels of TGF-βwere(14.32±1.25),(43.87±2.92),(28.33±3.11),(20.62±2.23)ng·L^-1.The levels of serum MCP-1,CRP,PGE2,TGF-βand relative expression of TLR4,MyD88 and NF-κB in the right carotid arteries in model group were significantly higher than those in sham-operation group,test group and control group were lower than those in model group,and those in contro
