摘　　要：目的:研究lncRNA?T199678基因对细胞生长周期的影响。方法:用lncRNA?T199678 smart silence转染SH?SY5Y细胞构建lncRNA?T199678沉默细胞株,用过表达lncRNA?T199678的质粒转染细胞构建lncRNA?T199678过表达细胞株,通过qPCR实验验证细胞内lncRNA?T199678的表达变化,利用流式细胞仪检测细胞生长周期变化。结果:qPCR验证发现lncRNA?T199678沉默组中细胞内lncRNA?T199678的表达明显低于NC对照组,lncRNA?T199678过表达组中细胞内lncRNA?T199678的表达明显高于NC对照组(P<0.05)。与对照组相比,lncRNA?T199678沉默组细胞处于G0/G1期的比例减少,S期的比例增加。与对照组相比,lncRNA?T199678过表达组细胞处于G0/G1期的比例增加,S期的比例减少。结论:靶向调控lncRNA?T199678基因可调控SHSY5Y细胞周期的进程。Objective:To investigate the effect oflncRNA?T199678 gene on cycle of SHSY5Y cells.Methods:lncRNA?T199678 smart silence was transfected into SHSY5Y cells to interfere lncRNA?T199678 expression.The lncRNA?T199678 expression plasmid was constructed and transfected into SHSY5Y cells.The expression of lncRNA?T199678 were detected by real?time PCR.Cell cyclewas detected by flow cytometry.Results:The results of RT?qPCR showed that the expression of lncRNA?T199678 in siT199678 group were significantly lower than that in NC group while the expression of lncRNA?T199678 in the recombinant plasmid group was significantly higher than that in NC group.The cells percentages in G0/G1 phases were decreased while that in S phase were increased after down?regulation of lncRNA?T199678 expression.On the contrary,after overexpression of gene lncRNA?T199678,the cells percentages in G0/G1 phases were increased while that in S phase were decreased.Conclusion:lncRNA?T199678 can interfere cell cycle progression of SHSY5Y cells.
