摘  要:目的探讨YAP1和SRPX2在非小细胞肺癌(non-small cell lung cancer,NSCLC)中的表达与微血管密度(microvessel density,MVD)及预后的关系。方法采用Western blot法检测NSCLC及其癌旁正常组织中YAP1和SRPX2的表达;采用免疫组化SP法检测NSCLC及其癌旁正常组织中YAP1、SRPX2和CD34的表达,并结合临床病理特征进行相关性分析。结果Western blot实验显示YAP1和SRPX2在NSCLC组织中的表达均高于癌旁正常组织(P<0.05)。免疫组化结果显示YAP1和SRPX2在NSCLC组织中均阳性,阳性率分别为62.5%(50/80)和55%(44/80);YAP1、SRPX2的表达与TNM分期、淋巴结转移相关(P<0.05)。YAP1和SRPX2阳性组的MVD值高于阴性组(P<0.05);NSCLC组织中YAP1和SRPX2表达水平均呈正相关(r=0.545,P<0.05),且YAP1和SRPX2表达与MVD值呈正相关。生存分析显示,YAP1和SRPX2阳性组生存期明显低于阴性组。结论YAP1和SRPX2在NSCLC中的表达具有相关性,两者可能在NSCLC的血管生成中发挥重要作用,联合检测YAP1和SRPX2的表达,可能为NSCLC的治疗提供新的预测指标和靶点。Purpose To investigate the expression of YAP1 and SRPX2 in non-small cell lung cancer(NSCLC)and the relationship between microvessel density(MVD)and prognosis.Methods Western blot was performed to assess YAP1 and SRPX2 expressions in NSCLC tissues and their adjacent tissues.Immunohistochemistry of SP was performed to assess YAP1,SRPX2 and CD34 expressions in NSCLC tissues and their adjacent tissues.Results Western blot showed that the expression levels of YAP1 and SRPX2 in NSCLC tissues were higher than that in the corresponding paracancerous tissue.Immunohistochemistry showed that YAP1 and SRPX2 were highly expressed in NSCLC tissues,and the positive expression rates were 62.5%(50/80)and 55%(44/80)respectively.YAP1 and SRPX2 expression was closely related to the TNM stage,lymph node metastasis(P<0.05).The MVD expression level of the YAP1 and SRPX2 positive expression group was higher than that in negative YAP1 and SRPX2 expression group(P<0.05).The expression level of YAP1 protein was significantly and positively correlated with that of SRPX2 protein(r=0.545,P<0.05).The expression level of YAP1 and SRPX2 was positively correlated with that of MVD.Survival analysis showed that the survival time of YAP1 and SRPX2 positive expression group was significantly lower than that in negative expression group.Conclusion The expression of YAP1 and SRPX2 is significantly correlated in NSCLC.YAP1 and SRPX2 may play crucial roles in the angiogenesis of NSCLC,which provide a new prognostic indicator and therapeutic targets in patients with NSCLC.
