摘 要:开发了一种高效检测小麦中交链孢酚单甲醚(alternariolmethyl ether,AME)的高效液相色谱(HPLC)检测方法。将样品采用乙腈/甲醇/水(45/10/45,体积比),30℃振荡提取30min后,经Bond Elut Plexa固相萃取小柱净化后,上样检测,检测器采用紫外检测器。该方法AME检测限为2.52μg/kg,定量限为8.40μg/kg,线性范围为0.25~2.00μg/mL,加标样品中不同浓度的AME回收率为80.1%~88.3%,日内精密度和日间精密度分别为2.13%~6.59%和3.57%~8.43%。In this study,a high-performance liquid chromatography(HPLC)method for the efficient detection of alternariolmethyl ether(AME)in wheat was developed.The sample was extracted with acetonitrile/methanol/water(volume ratio 45/10/45),shaken at 30℃for 30 minutes,and purified by Bond Elut Plexa solid-phase extraction cartridge.Then the sample was detected with a Variable wavelength detector.In this method,the limit of detection(LOD)of AME was 2.52μg/kg,and the limit of quantification(LOQ)was 8.40μg/kg,and the linear range was 0.25~2.00μg/mL.The concentration of AME recovery was 80.1%~88.3%,the intra-day precision and inter-day precision were 2.13%~6.59%and 3.57%~8.43%,respectively.
