摘  要:目的探讨酪氨酸蛋白激酶1(tyrosine-protein kinase 1,STYK1)在肺癌组织中的表达水平、对肺癌细胞生存的影响及其作用机制。方法定量PCR检测STYK1在35例肺癌组织中的表达水平,分析其与患者临床病理特征间的关系。沉默肺癌细胞A549和HCC827中STYK1基因,定量PCR检测mRNA表达水平,Western blot检测蛋白表达水平,CCK-8法检测细胞增殖,流式细胞术检测细胞凋亡。结果STYK1在肺癌组织中表达水平升高(P<0.01)。其表达水平在肺癌病理分期方面比较,差异具有统计学意义(P<0.01)。沉默STYK1后,肺癌细胞增殖活性降低,细胞凋亡率升高,差异均具有统计学意义(均P<0.01)。沉默STYK1抑制Akt磷酸化,下调Bcl-2和Bcl-xL表达水平(均P<0.01)。结论STYK1在肺癌组织中呈高表达。其可能通过激活Akt信号通路,促进肺癌细胞增殖。Objective To investigate the expression of STYK1 in lung cancer tissues,and study the effects and mechanism of STYK1 on the lung cancer cell proliferation.Methods The mRNA expression of STYK1 in 35 paired lung cancer tissues and paracancer tissues was detected by qPCR.The relationship among the expression of STYK1 and the clinicopathological characteristics were analyzed.siRNA was applied to silence the expression of STYK1 in A549 and HCC827 cells.The mRNA and protein expressions were detected by qPCR and Western blot,respectively.CCK-8 assay was used to determine the cell viability,and flow cytometry was used to detect the cell apoptosis.Results The expression of STYK1 in lung cancer tissues was significantly higher than that in the paracancer tissues(P<0.01).In addition,the expression of STYK1 was positively correlated with the progression of lung cancer(P<0.01).Moreover,knockdown of STYK1 inhibited cell proliferation,and promoted cell apoptosis in lung cancer cells(both P<0.01).Knockdown of STYK1 attenuated the phosphorylation of Akt,followed by decreased expressions of Bcl-2 and Bcl-xL(all P<0.01).Conclusions The expression of STYK1 is significantly increased in lung cancer tissues,which promotes the proliferation of lung cancer cell via activating Akt signal pathway.
