摘 要:目的观察丰富环境(enriched environment,EE)对大鼠脑缺血再灌注(cerebral ischemia reperfusion,CIR)损伤后血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法将成年雄性SD大鼠64只随机分为正常组、假手术组、CIR模型组和EE干预组,每组16只。采用Longa法建立大鼠大脑中动脉缺血再灌注模型。EE干预组大鼠给予EE饲养,其余大鼠均按照标准环境饲养,共饲养14 d。于EE干预的第1天及第14天观察大鼠脑缺血半暗带组织病理形态学改变;采用实时荧光定量PCR(qRT-PCR)检测大鼠脑缺血半暗带VEGF mRNA表达;采用免疫组化检测大鼠脑缺血半暗带VEGF蛋白表达。结果EE干预的第1天,正常组与假手术组大鼠脑组织结构清晰,神经元排列规则,细胞核仁清晰,CIR模型组与EE干预组大鼠脑缺血半暗带均出现水肿、神经元脱失等病理改变;CIR模型组和EE干预组大鼠脑缺血半暗带VEGF mRNA及其蛋白表达均高于假手术组和正常组(均P<0.05),而CIR模型组与EE干预组比较差异无统计学意义(P>0.05)。EE干预的第14天,CIR模型组与EE干预组脑缺血半暗带水肿均减轻,且EE干预组改善稍明显;EE干预组大鼠脑缺血半暗带VEGF mRNA及其蛋白表达高于CIR模型组(均P<0.05)。结论EE可能通过增加内源性VEGF表达这一途径改善大鼠CIR损伤。Objective To observe the effect of enriched environment(EE)on the expression of vascular endothelial growth factor(VEGF)after cerebral ischemia reperfusion(CIR)injury in rats.Methods 64 adult male SD rats were randomly divided into four groups:a normal group,a sham group,a CIR model group,and an EE intervention group,with 16 rats in each group.Longa method was used to establish the rat middle cerebral artery ischemia-reperfusion model.The rats in the EE intervention group were fed with EE,and the other rats were fed in accordance with the standard environment.On the first day and the 14th day of EE intervention,the ischemic penumbra tissues of rats in each group were taken to observe the pathomorphological changes.Quantitative real-time PCR(qRT-PCR)was used to detect the expression of ischemic penumbra VEGF mRNA in rats in each group.Immunohistochemistry was used to detect VEGF protein expression in the ischemic penumbra of rats in each group.Results On the first day of EE intervention,HE staining showed clear brain structure,regular arrangement of neurons and clear cell nucleoli of rats in the normal group and the sham group.The cerebral ischemia penumbra of rats in the model group and the EE intervention group showed edema,neuronal prolapse and other pathological changes.The expression of VEGF mRNA and protein of rats in the model group and the EE intervention group were higher than those in the sham group and the normal group(all P<0.05),while the difference between the model group and the EE intervention group was not statistically significant(P>0.05).On the 14th day of EE intervention,HE staining showed that the cerebral tissue edema decreased in both the cerebral ischemia-reperfusion model group and the EE intervention group,and the improvement was more obvious in the EE intervention group.The expression of VEGF mRNA and protein of rats in EE intervention group were higher than those in the ischemic reperfusion model group(all P<0.05).Conclusions The EE may improve the CIR injury in rats by increasing the e
