子宫内膜癌组织Runx3、Twist1表达与临床病理特点及预后的关系

摘　　要：目的探讨子宫内膜癌(EC)组织中RUNX家族转录因子3(Runx3)、Twist家族碱性螺旋-环-螺旋转录因子1(Twist1)表达与临床病理参数和预后的关系。方法选择80例EC患者,通过荧光定量PCR法检测子宫内膜癌组织及癌旁组织中Runx3、Twist1表达,比较两种组织Runx3、Twist1表达量,并对Runx3、Twist1表达与子宫内膜癌患者临床病理学特征之间的关系进行分析,采用Kaplan-Meier法分析不同Runx3、Twist1表达与患者预后生存的关系。结果与癌旁组织相比,EC组织中Runx3表达降低,Twist1表达增高(P均<0.05)。EC组织中Runx3和Twist1表达呈负相关(r=-0.611,P<0.05)。EC组织中Runx3、Twist1表达与肿瘤临床分期、肿瘤分化程度有关(P均<0.05),而与年龄、病理类型、肿瘤大小、肌层浸润深度及淋巴结转移无关(P均>0.05)。低Runx3表达组3年生存率低于高Runx3表达组,高Twist1表达组3年生存率低于低Twist1表达组(P均<0.05)。结论EC中Runx3表达下调,Twist1表达上调,两者表达与肿瘤分期、肿瘤分化程度及预后有关。Objective To investigate the expression of RUNX family transcription factor 3(Runx3)and Twist family basic helix-loop-helix transcription factor 1(Twist1)in the endometrial cancer(EC)tissues and its relationship with clinicopathologic characteristics and prognosis.Methods Eighty EC patients were selected and the expression levels of Runx3 and Twist1 in EC and adjacent tissues were detected by fluorescence quantitative PCR.The expression levels of Runx3 and Twist1 in the two tissues were compared,and the relationships of the expression levels of Runx3 and Twist1 with the clinicopathologic characteristics of EC patients were analyzed.Kaplan-Meier method was used to analyze the relationship between Runx3 and Twist1 expression and prognosis of patients.Results Compared with the adjacent tissues,the expression of Runx3 decreased and the expression of Twist1 increased in the EC tissues(both P<0.05).There was a negative correlation between Runx3 and Twist1 expression in EC tissues(P<0.05).The expression of Runx3 and Twist1 in the EC tissues was related to the clinical stage of tumor and the degree of tumor differentiation(both P<0.05),but not related to age,pathological type,tumor size,depth of myometrial invasion,or lymph node metastasis(all P>0.05).The 3-year survival rate of patients with low Runx3 expression was significantly lower than that of patients with high Runx3 expression,and 3-year survival rate of patients with high Twist1 expression was significantly lower than that of patients with low Twist1 expression(all P<0.05).Conclusion The expression of Runx3 in EC tissues is down-regulated,while the expression of Twist1 is up-regulated;both of them are related to the tumor stage,tumor differentiation and prognosis of patients.
