摘  要:目的探讨长链非编码RNA ASB16-AS1(LncRNA ASB16-AS1)对前列腺癌细胞增殖、迁移、侵袭的影响及其分子机制。方法体外培养前列腺癌细胞株LNCaP,分别将si-NC、si-ASB16-AS1、miR-NC、miR-760 mimics、si-ASB16-AS1+anti-miR-NC、si-ASB16-AS1+anti-miR-760转染至LNCaP细胞。采用MTT检测细胞增殖能力,Transwell小室实验检测细胞迁移及侵袭能力,双荧光素酶报告实验验证ASB16-AS1与miR-760的靶向关系,Western blotting法检测细胞周期蛋白1(CyclinD1)、基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)、p21蛋白水平。结果与转染si-NC、miR-NC相比,转染si-ASB16-AS1、miR-760 mimics后细胞增殖活力降低(P均<0.05),迁移、侵袭细胞数减少(P均<0.05)。双荧光素酶报告实验证实ASB16-AS1能够靶向结合miR-760。与si-ASB16-AS1+anti-miR-NC组相比,si-ASB16-AS1+anti-miR-760组细胞增殖活力升高(P<0.05),迁移、侵袭细胞数增多(P均<0.05),CyclinD1、MMP-2、MMP-9蛋白水平升高(P均<0.05),p21蛋白水平降低(P<0.05)。结论LncRNA ASB16-AS1通过靶向干扰miR-760表达从而促进前列腺癌细胞增殖、迁移及侵袭。Objective To investigate the effects of LncRNA ASB16-AS1 on the proliferation,migration,and invasion of prostate cancer cells and its molecular mechanism.Methods The prostate cancer cell line LNCaP was cultured in vitro,and si-NC,si-ASB16-AS1,miR-NC,miR-760 mimics,si-ASB16-AS1 and anti-miR-NC,si-ASB16-AS1 and anti-miR-760 were transfected into LNCaP cells.MTT was used to detect the cell proliferation.Transwell chamber experiment was performed to detect cell migration and invasion abilities.Dual luciferase report experiment verified the targeting relationship between ASB16-AS1 and miR-760.Western blotting was used to detect the protein levels of CyclinD1,matrix metalloproteinase-2(MMP-2),MMP-9,and p21 protein.Results Compared with si-NC and miR-NC transfection,the cell proliferation activity decreased after transfection of si-ASB16-AS1 and miR-760 mimics(P<0.05),and the number of migrating cells and invading cells was significantly reduced(both P<0.05).Dual luciferase reporting experiment confirmed that ASB16-AS1 could bind to miR-760.Compared with the si-ASB16-AS1+anti-miR-NC group,the cell proliferation activity of the si-ASB16-AS1+anti-miR-760 group increased(P<0.05),and the number of migrating cells and invading cells increased significantly(both P<0.05),the protein levels of CyclinD1,MMP-2,and MMP-9 significantly increased(all P<0.05),and the protein level of p21 decreased(P<0.05).Conclusion LncRNA ASB16-AS1 promotes the proliferation,migration and invasion of prostate cancer cells by interfering the miR-760 expression.
