摘  要:杨叶甲是典型的食叶的叶甲昆虫,主要取食杨树叶,是重要的林业经济害虫。目前有关杨叶甲,乃至有关叶甲的形态学研究都十分少见。本文以杨叶甲成虫为样本,利用显微CT技术获取其头部的三维形态数据。样本经酒精梯度脱水和临界点干燥处理后,将其置于X-射线微型计算机断层扫描仪(Skyscan 1172)的样本台上,在光波电子束强度为40 keV下对其头部进行扫描。扫描获得1554张杨叶甲成虫头部断层二维图像序列,存储总容量为18.7 GB,图像空间分辨率为1.04μm。断层图图像质量清晰,无环形伪影,样本杨叶甲头部内部组织结构也清晰完整,肌肉、骨骼、消化道、甚至神经系统清晰可见。基于图像序列可对杨叶甲成虫头部内、外部形态结构进行三维重建,为甲虫头部形态学、解剖学的研究提供基础数据源。Chrysomela populi is a typical leaf beetle feeding on mainly leaves of aspen trees,and a major pest for forestry production.Morphological studies on Chrysomela populi,or even on chrysomelid beetles,are surprisingly scarce.In this investigation,Chrysomela populi adults were used for x-ray micro-computed tomography(micro-CT).Before they were scanned with a Skyscan 1172 micro-CT(beam strength:40 keV),the adult samples were dehydrated with ethanol and dried to the critical point.The scanned image sequence stack includes 1,554 images with a resolution of 1.04μm,totaling a data volume of 18.7 GB.The image quality is clear and there are no annular pseudomonas.The tissue in the head is also clear and intact;muscles,bones,digestive tracts,and even nervous systems are clearly visible.The scanned image sequence stack could be used for 3D reconstruction and anatomical study of the adult head of Chrysomela populi.This dataset provides basic data for the study of beetle head morphology and anatomy.
