摘 要:目的:体外研究一种具有靶向性、副作用少、同时具备诊断和治疗作用的针对头颈鳞癌治疗的微泡颗粒。方法:分别制备载吉非替尼脂质体-微泡模型和载氯喹脂质体-微泡模型。将HN-30细胞分为5组,即PBS缓冲液组、载吉非替尼脂质体+载氯喹脂质体(GE-Lipo+CQ-Lipo)组、载吉非替尼脂质体-微泡模型(MB-GE-Lipo)组、载氯喹脂质体-微泡模型(MB-CQ-Lipo)组和载吉非替尼脂质体-微泡模型+载氯喹脂质体-微泡模型(MB-GE-Lipo+MB-CQLipo)组。检测各组细胞的增殖抑制效果。采用SPSS 17.0软件包对数据进行统计学分析。结果:当卵磷脂酰胆碱(egg phosphatidylcholine)∶胆固醇(Cholesterol)=2∶1时,与8∶1组相比,包封率无显著差异(P>0.05),2∶1与5∶1、10∶1、20∶1组相比,差异有统计学意义(P<0.05);5∶1、8∶1、10∶1、20∶14组之间两两比较,无显著差异(P>0.05)。对于HN-30细胞的增殖抑制作用,(MB-GE-Lipo+MB-CQ-Lipo)组与PBS组相比,差异有统计学意义(P<0.05);(MB-GE-Lipo+MB-CQ-Lipo)组与(MB-GE-Lipo)组相比,差异无统计学意义(P>0.05)。结论:当EPC∶Chol=5∶1时,载吉非替尼脂质体包封率较高,且胆固醇比例合适,脂质体较稳定。MB-GE-Lipo+MB-CQ-Lipo联合超声对HN-30细胞的增殖抑制作用明显。PURPOSE: To explore a kind of therapeutic particle of targeted, fewer side effects, both with value of diagnosis and treatment for head and neck squamous cell carcinoma(HNSCC) in vitro. METHODS: Gefitinib-liposomemicrobubble model and chloroquine liposome-microbubble model were prepared separately. HN-30 cells were divided into 5 groups: PBS, GE-Lipo+CQ-Lipo, MB-GE-Lipo, MB-CQ-Lipo, MB-GE-Lipo+ MB-CQ-Lipo. MTS kit was used to detect the inhibitory effect of cell proliferation in each group. SPSS17.0 software package was used to analyze the data.RESULTS: When EPC: Chol=2:1, there was no significant difference in the entrapment efficiency between 2:1 and 8:1 group(P>0.05). Compared with 5:1,10:1,20:1 group, there was significant difference in the entrapment efficiency of 2:1 group(P<0.05). Among group 5:1, 8:1, 10:1, and 20:1, compared with each other, there was no significant difference( P>0.05). For HN-30 cells, there was significant difference(P<0.05) between MB-GE-Lipo+ MB-CQ-Lipo group and PBS group. There was no significant difference between MB-GE-Lipo+ MB-CQ-Lipo group and MB-GE-Lipo group(P>0.05).CONCLUSIONS: When EPC:Chol=5:1, the EE of GE-Lipo was higher, the ratio of cholesterol was appropriate, and the liposome was more stable. The inhibitory effect of gefitinib liposome-microbubbles + chloroquine liposome-microbubbles combined with ultrasound on HN-30 cell proliferation was significant.
